Metabolics Pharma Announces ENT-03 Generates Sustained Weight Loss and Liver Pathology Improvements in an Animal Model for Obesity

DOYLESTOWN, Pa.--(BUSINESS WIRE)--Metabolics Pharma ('Metabolics'), a privately held, Doylestown-based, clinical-stage biopharmaceutical company pioneering novel treatments for metabolic diseases, today announced that its drug candidate, ENT-03, showed significant benefit in metabolic dysfunction in addition to sustaining weight loss, improvement in blood glucose, and insulin sensitivity in comparison to semaglutide in an animal model for obesity.

'These preclinical data highlight both the promise and potential value of ENT-03 in the treatment of metabolic disease. The research shows that ENT-03 clearly improves insulin sensitivity and sustained weight reduction that persists following cessation of treatment. This research is the foundation for our Phase 1 clinical trials in healthy obese and diabetic obese subjects.'
ENT-03 is a novel, centrally acting aminosterol with Protein Tyrosine Phosphatase 1B (PTP1B) inhibitory activity, which normalizes glucose and causes weight loss by acting on brain circuits that regulate energy and metabolism. In obese, diabetic mice, ENT-03 rapidly lowers blood glucose, reduces food intake and adiposity, eliminates liver fat, and improves liver function.
This study compared ENT-03 to semaglutide, a combination of the two compounds, and vehicle in Diet Induced Obesity (DIO) mice. While the vehicle-treated mice gained weight through the duration of the study, mice treated with ENT-03, Semaglutide, or a combination of the two drugs all lost weight and reduced insulin and glucose levels. ENT-03 showed a greater reduction in weight than Semaglutide over the 10-week treatment period. Additive weigh loss was seen when the drugs were used in combination. After cessation of therapy, weight loss and reduction in glucose and insulin were sustained for an additional two months in the ENT-03 treated mice but not Semaglutide treated mice. In addition, ENT-03 showed substantially greater preservation of lean mass in comparison to other treatment groups.
'GLP-1 agonists have revolutionized the treatment of obesity and type 2 diabetes,' said Richard Larson, MD, PhD. Metabolics Pharma has completed a Phase 1a, first in human, randomized, double-blind, single ascending dose study of ENT-03. Results of the Phase 1a trial were presented at the American Diabetes Association 85 th Scientific Sessions on June 22, 2025. A Phase 1b clinical trial is planned to initiate in the second half of 2025.

Lululemon Athletica Canada Inc. is accusing Costco Wholesale Corp. of infringing on its intellectual property by selling knockoffs of some of its most popular products. A lawsuit filed in a California court recently alleges Costco sells dupes of Lululemon's Scuba hoodies and sweatshirts, Define jackets and ABC pants. Some of the alleged fakes Vancouver-based Lululemon identified are sold under Costco's private label Kirkland, but others are made by manufacturers Danskin, Jockey and Spyder. 'Some customers incorrectly believe these infringing products are authentic Lululemon apparel while still other customers specifically purchase the infringing products because they are difficult to distinguish from authentic Lululemon products, particularly for downstream purchasers or observers,' the lawsuit says. Lululemon alleges these scenarios take advantage of patents it holds, as well as the reputation and goodwill it has built with customers. In its 49-page court filing, Lululemon says it tried to address the dupes by sending Costco cease and desist letters but is now asking a court to step in. Lululemon has requested the matter be heard by a jury trial, which it wants to order Costco to cease manufacturing, importing, marketing and selling the alleged dupes. It also wants the Washington-based retailer to remove any instances where it was advertising alleged dupes online or in print and is asking the court to require Costco to cover any lost profits Lululemon incurred from the products. Costco did not immediately respond to a request for comment. It has yet to file a response to Lululemon's lawsuit. Dupes have become incredibly popular in recent years as shoppers looked to cope with inflation and a global trade war by seeking products mimicking the originals made by name brands. The phenomenon has gained traction online in particular, with social media accounts sharing the best dupes they've found. While knock-off cosmetics are especially popular, Lululemon has also become a target because it sells its athletic wear at higher prices, which some customers find unattainable. The lawsuit comes weeks after Lululemon said it would try to weather new and possibly incoming tariffs with price increases that will be 'modest in nature' and applied only to a 'small' portion of its products. The brand has been working to bounce back from a lack of newness that disappointed consumers last year and earlier this year. One great option Guild has for lower-income homebuyers is the 1% Down mortgage. Borrowers who meet income requirements and have a 1% down payment are eligible for an additional 2% from Guild in the form of a non-repayable grant, up to $5,000. Unlike similar programs from competitors, the 1% Down loan is available to both first-time and repeat buyers with no location restrictions. Apply online for rates. Warner predicts Trump's "Big Beautiful Bill" will cripple GOP support

Senate Republicans released updated megabill text late Friday that would make sharp cuts to the Inflation Reduction Act's solar and wind tax credits after a late-stage push by President Donald Trump to crack down further on the incentives. The text would require solar and wind generation projects seeking to qualify for the law's clean electricity production and investment tax credits to be placed in service by the end of 2027 — significantly more restrictive than an earlier proposal by the Senate Finance Committee that tied eligibility to when a project begins construction. The changes came after Trump urged Senate Majority Leader John Thune to crack down on the wind and solar credits and align the measure more closely with reconciliation text, H.R.1, that passed the House, as POLITICO reported earlier on Friday. The changes are likely to put some moderate GOP senators, who have backed a slower schedule for sunsetting those incentives, in a tough position. They'll be forced to choose between rejecting Trump's agenda or allowing the gutting of tax credits that could lead to canceled projects and job losses in their states — something renewable energy advocates are also warning about. 'We are literally going to have not enough electricity because Trump is killing solar. It's that serious,' Sen. Brian Schatz (D-Hawaii) responded on X early Saturday. 'We need a bunch of new power on the grid, and nothing is as available as solar. Everything else takes a while. Meantime, expect shortages and high prices. Stupid.' The revised text would retain the investment and production tax credits for baseload sources, such as nuclear, geothermal, hydropower or energy storage, as proposed in the Finance Committee's earlier proposal. But it would make other significant changes, including extending a tax credit for clean hydrogen production until 2028. The panel's earlier proposal would have eliminated the credit after this year. And despite vocal lobbying by the solar industry, the proposal would maintain an abrupt cut to the tax incentive supporting residential solar power. The committee's earlier proposal would have eliminated that credit six months after the enactment of the bill; now the updated draft proposes repealing it at the end of this year. It would also deny certain wind and solar leasing arrangements from accessing the climate law's clean electricity investment and production tax credits, but, in a notable change, removed earlier language specifically disallowing rooftop solar. And it would move up the timeline for certain rules barring foreign entities of concern from accessing those credits. The bill would move up the termination date for electric vehicle tax credits to Sept. 30, compared to six months after enactment in the earlier Finance text. The credit for EV chargers would extend through June 2026. The new text also provides a bonus incentive for advanced nuclear facilities built in communities with high levels of employment in the nuclear industry. And the bill makes metallurgical coal eligible for the advanced manufacturing production tax credit through 2029. Sam Ricketts, co-founder of S2 Strategies, a clean energy policy consulting group, said the new draft is going to 'screw' ratepayers, kill jobs and undermine U.S. economic competitiveness. 'All just to give fossil fuel executives more profits,' he said. 'Or to own the libs. Insanity.' Josh Siegel contributed to this report.
