Harris, the Paris Olympics silver medallist, powered home in 24.02sec to beat Chinese pair Wu Qingfeng (24.26) and Cheng Yujie (24.28).
The American Walsh, who won gold in the 50m and 100m butterfly in Singapore, finished fourth in 24.40.
Australia also won the men's 50m freestyle, with Cam McEvoy taking gold on Saturday.
Harris said becoming an individual world champion was "the dream I have been dreaming of the whole time".
"This is why I swim," said the 23-year-old, who could not stop smiling as the magnitude of her achievement set in.
"I still can't put it into words, it'll take a while to process."
World record holder Sarah Sjoestroem of Sweden, the reigning Olympic champion and a four-time world title-holder in the event, did not compete in Singapore.
"I love sprinting, I love racing. Could not be more happy," added Harris.
The Australian has now won six world golds in her career. The previous five were in relay events.

AFC hit China's Shandong with two-year ban over no-show - World
China's Shandong Taishan have been banned from Asian club competitions for two years and fined after their no-show for a crunch match earlier this year. The Chinese Super League side were booted out of the region's Champions League Elite in February after withdrawing hours before a game against South Korea's Ulsan. The Asian Football Confederation's (AFC) disciplinary and ethics committee has now barred the team from all AFC tournaments "up to and including the 2026/27 season", according to a summary of a July 30 decision posted online on Monday. It fined the club $50,000. Shandong was ordered to repay a $600,000 participation fee and a $200,000 performance bonus to the Kuala Lumpur-based AFC. It must also pay compensation of $40,000 to South Korea's Ulsan for "damages and losses claimed". After withdrawing at the 11th hour from the February 19 match, Shandong said they were "unable to form a team" because of "serious" health issues within the travelling squad. "We deeply regret this situation and apologise to the AFC, Ulsan FC, the fans, and the community," they said at the time, without explaining exactly what the health issues were. Ghana's Partey granted bail on rape charges - Africa
Former Arsenal footballer Thomas Partey was granted conditional bail on Tuesday after appearing in a London court on rape and sexual assault charges. The 32-year-old Ghana international has been charged with five counts of rape against two women, as well as a charge of sexual assault against a third woman. The alleged offences took place between 2021 and 2022, when he was an Arsenal player. Partey was charged four days after leaving the Gunners when his contract expired at the end of June. The midfielder stood with his arms behind his back in the dock at Westminster Magistrates' Court, wearing a black zip-neck jumper. British police said they first received reports of an allegation of rape in February 2022. Partey's lawyer Jenny Wiltshire previously said he "denies all the charges against him", adding he welcomed "the opportunity to finally clear his name". Partey joined Arsenal for £45 million ($60 million) from Atletico Madrid in October 2020. He appeared 52 times for the Gunners last season, scoring four goals. Overall he played 167 games for the club, scoring nine times. Partey has also made 51 appearances for Ghana. An Arsenal spokesman previously said: "The player's contract ended on June 30. Due to ongoing legal proceedings the club is unable to comment on the case." Reports in recent days have linked Partey with a move to Spanish club Villarreal. Israa Farhan Former US President Donald Trump has signed an executive order to establish a White House Olympic Task Force in preparation for the Los Angeles 2028 Summer Olympics. The task force will focus on federal coordination across security, infrastructure, transportation, and visa-related issues. According to a senior official, the team will include members from Trump's administration and various federal agencies. Their primary role will be to streamline efforts between the federal government, states, and local authorities in the lead-up to the Games. The order also aims to simplify visa and accreditation procedures for international athletes, coaches, officials, and media representatives, ensuring smoother entry and operations during the event. The Los Angeles Summer Olympics are scheduled for 2028, just three years away. Trump, who lost the 2020 election to Joe Biden, has expressed enthusiasm about potentially overseeing the Olympics during what he refers to as his second term. The event will follow another major sports milestone, the 2026 FIFA World Cup, which the US is co-hosting.
