Hair loss may seem like a losing battle, but with the right products, it's totally possible to regain and regrow thinning hair. For the record, hair loss is extremely common in men, and most would (naturally!) rather keep their youthful strands and slow down or even reverse thinning and balding. If you're tired of dealing with the embarrassment of hair loss, why not take matters into your own hands and try a few new products?Dr. Amy Forman Taub, a dermatologist and an assistant professor at Northwestern University Medical School, tells Yahoo Life, "You can effectively stop genetic hair loss to a large degree." But it can be harder to reverse prolonged genetic hair loss and grow back the hair you've lost, she adds, so early intervention is important. Whether your hair loss is due to genetics, COVID, stress, poor diet or illness, you can restore hair health with the right products and a little patience. In order to help you find the best hair growth products to suit your needs and increase your odds of success, we consulted with dermatologists and hair loss specialists to identify the key ingredients to look for in hair-regrowth products. And because every person — and every budget — is different, we made sure to include a range of products and price points to ensure that there's something for everyone. Read more: Looking for more ways to regrow thinning or sparse hair? We've reviewed the best vitamins and supplements for hair growth, the best hair growth oils and serums and the best shampoos for thinning hair. We even dedicated a whole section to the best hair growth products for women, according to dermatologists.

Dr. Michelle Henry, a board-certified dermatologist specializing in hair loss, tells Yahoo Life, "It's important to first consider the root cause of your hair loss." There are several different types of hair loss (scroll down for more on that), and the best hair growth product for you is one that addresses that cause.
There are two FDA-approved ingredients used to target hereditary hair loss, which is the most common form of hair loss in men: minoxidil and finasteride. "These treatments have demonstrated positive results in stimulating hair regrowth," Dr. Henry explains.
You'll also want to choose a hair loss product you know you can continue to use. "If your loss is due to genetics, you must remain on medication or treatments indefinitely, otherwise your hair loss will revert to the point where it would be without treatment," Taub says.
Type of hair loss: Hair loss in men is often due to genetics, according to the American Academy of Dermatology (AAD). This is called male-pattern hair loss or androgenic alopecia. If you have this condition, it means that you've inherited genes that cause your hair follicles to shrink and eventually die, the AAD explains. While this shrinking can start as early as your teens, it usually starts later in life. But that's not the only potential cause of hair loss in men. In addition to genetics, COVID-19 and the flu can cause temporary hair loss, as can stress and poor diet. If you're unsure of what could be behind your hair loss, it's a good idea to consult with a dermatologist.
Type of hair loss products: You have options when it comes to choosing a hair growth product for men. These products come in a range of forms, such as serums, shampoos, oils, foams, prescription medications, light therapy, micro-needling, vitamins and supplements. It's important to choose a product you like and will continue using, and you may want to use some products in conjunction — for instance, a shampoo and a supplement. Just be sure to talk to your dermatologist to make sure the course of action you're taking makes sense for your type of hair loss.
Budget: Hair growth products for men are an investment and, again, you'll need to use them consistently to see the best results. With that, it's important to determine how much you feel comfortable spending on a regular basis and choose a treatment from there. While there are a range of price points on the market, doctors say spending more isn't necessarily better. Instead, it's important to choose the right product for you with the key ingredients that address your condition.
Prescription vs. nonprescription hair growth treatment: If your hair loss is extreme, it's a good idea to consult with a board-certified dermatologist for guidance. They can help you decide on the right treatment for you, which may include prescription oral medications like finasteride (Propecia), spironolactone (CaroSpir, Aldactone) or dutasteride (Avodart).

To find the best hair growth products for men, we spoke with multiple dermatologists and hair loss experts for their input on key ingredients to look for and if one therapy or treatment was superior. Then we researched over 40 hair regrowth products for men, rating each based on use of the highest quality ingredients, easy application methods, safety, success rate and reviews from firsthand users.

Many factors go into whether a hair growth product will work for you, including the cause of your hair loss, how advanced it is and which products you choose. Not every hair growth product works for everyone, so you may need to do some trial and error to find one that's comfortable and effective. However, dermatologists say that these products typically will deliver good results — and they're constantly getting better. "We can expect hair loss to one day be a thing of the past," says Dr. Glenn Vallecillos, a board-certified plastic surgeon focusing on hair loss.
Using a hair growth product or treatment like a derma roller or light therapy cap can all help promote hair growth (consult with your dermatologist if you're unsure which one is right for you). It's important to start sooner rather than later since these products typically work best when you intervene early. At the same time, dermatologist Jose Diego Mier tells Yahoo Life that "you can promote good hair from the inside out. That means eating a balanced diet with plenty of fruits, vegetables and proteins to give your body the nutrients it needs to support hair health and growth." It's also a good idea to stay hydrated and do your best to manage your stress levels, he says.
Every person — and every product — is different. However, most hair growth and hair-thinning products say that users should notice a change in six to eight weeks. Again, using products sooner is your best bet for success.
It's hard to give a blanket recommendation, given that every person's needs are different. However, a product with a 5% minoxidil solution, or prescription Finsterade, has been leading the charge. It's often considered a top-tier treatment that works for most men. If you're unsure, consult a board-certified dermatologist who specializes in hair loss. They should be able to help steer you toward the best treatment for you.

The best hair growth oils and serums for 2025, tested and reviewed

There are hair oils to tame flyaways. Oils to finish your style with a super high shine. Hair oils formulated to help restore thinning hair and promote new growth. But how in the world do you know which product will help with what, and more to the point, choose the best hair oil when there are hundreds to choose from?We get it. The confusion is real. For starters, it's essential to consider your needs, hair type and skin sensitivity. For instance, those with sensitive skin should "opt for natural and nourishing products that are suitable for your hair type," said board-certified dermatologist Dr. Jose Diego Mier. Or maybe your skin is as tough as nails, in which case you want to opt for a powerful medicated solution like the FDA-approved hair growth treatment minoxidil. "The oldest established remedy and still one that is used quite a bit is topical minoxidil," said board-certified dermatologist Dr. Amy Forman Taub. Obviously, though, it is a major medication and has potential side effects that need to be evaluated for each person, she added. So for those looking for oils and serums to promote a healthy scalp and hair growth, we did the research. We spoke with six board-certified dermatologists and trichologists to learn what key ingredients to look for (and what ingredients to avoid). We then took our experts' feedback and researched what seemed like an endless range of products, then narrowed down the list of our favorites according to key ingredients, brand reputation, clinical studies, user feedback, success rates and cost. Our final list comprises the best hair-growth oils and serums for hair-thinning and hair-loss needs. The best hair growth oils and serums of 2025 What should you look for when shopping for the best hair growth oils? How we chose the best hair growth oils and serums FAQs Meet our experts Update, July 8, 2025: We checked all product prices and availability. Our No. 1 pick for best hair growth oils remains unchanged and is on sale now for Amazon Prime Day. (back to top) Consider your hair health needs: Most hair oils and serums can boost the length and fullness of your hair, but they can have other benefits, too. Some can help reduce scalp irritation, enhance growth or boost shine. Choose a product that meets all your needs. It's also important to consider if the product is formulated for a gender. The best hair products for women will not always match the best hair growth products for men. Key ingredients: There are two routes you can take: all-natural or chemical formulations. The all-natural route works best when the formulation includes rosemary, mint, peptides, Ayurvedic herbs, castor oil, biotin, fatty acids, tea tree, bergamot, Ashwagandha and pea peptides. You could also opt for one of three FDA-approved products: minoxidil, finasteride or baricitinib. But keep in mind that medical solutions will have side effects that you should discuss with a prescribing physician before trying. Clinically proven: A multitude of clinical studies on hair growth oils and serums are available online. If you find a product or ingredient you like, make sure it is backed by a clinical study that shows the ability of the product to deliver the results you want. Cost: It is important to keep in mind that most products require at least three months of use, so make sure you can afford to keep buying the product for a few months. You could also opt to subscribe and save, which is available on many brand and retail websites. Type: The consistency of the hair oil or serum will play a part in how well it will work for your hair and how it will affect the look of your hair. A heavy oil will likely be too much for thin hair. However, a lightweight oil or serum may not work well or be moisturizing enough for coarse or curly hair. Note: If you find that topical solutions are too heavy, even the lightest-weight solutions, you may benefit from a vitamin or supplement for hair growth. (back to top) First, we consulted with experts, including board-certified dermatologists, trichologists and plastic surgeons. We then reviewed numerous hair oils and serums that were specifically formulated to promote thicker, fuller hair and growth. We then narrowed down the list according to key ingredients, clinical studies, in-house tester and customer satisfaction, brand reputation and consistency. While the best hair oils and serums for hair growth depend on your specific concerns and goals, we found that cost and effectiveness tend to be favored over extraneous features like scent and packaging. Keeping the different reasons you may want hair oil and the range of hair types in mind, we opted to include a broad range of products, from scalp serums to root-to-tip oils, which included both budget-friendly options and pricier products. We strived not to exclude any company based on its price point. Still, to make our list, it had to have enough positive points from consumers, well-researched ingredients, and hair-and-scalp benefits to be worth its cost. (back to top) Dr. Andy Goren, a board-certified dermatologist, trichologist and professor said: "There are several common causes of hair loss like alopecia, high cortisol levels, stress, postpartum, hair shedding, etc. — genetic predisposition happens to be the most common, specifically hormonal and genetic-mediated alopecia (referred to as androgenetic alopecia or pattern hair loss). Other forms include immune-mediated alopecia (such as alopecia areata), traction alopecia (such as from hair extensions) and telogen effluvium often caused by stress or illness (COVID hair loss is absolutely a thing!). Finally, another form of alopecia is chemotherapy-induced alopecia caused by chemotherapy." (back to top) As a rule, a hair serum will be less oily and better for fine hair, while an oil will be on the heavier side but will work wonders for coarse or curly hair. That all said, before you discount an oil, it can offer extra benefits many serums cannot, such as adding a high shine to a finished style and taming flyaways; also, hair growth oils are almost always fortified with hair and scalp helpers, such as biotin, nettle, peptides, mint and rosemary oil. How do hair growth oils and serums work? Many of the best hair growth products are multifaceted and offer various benefits that work together to promote hair health (and, hopefully, improve density and length as well). Many rely on plant-based formulas (though some use synthetics) to reduce inflammation, exfoliate and stimulate the scalp, protect the hair follicle and shaft, and nourish it from root to tip. How often should I use hair growth oils? Most products give you precise application instructions along with a timeline. While the only really issue with prolonged hair oil and serum use is buildup, once you reach the point where your hair health is optimal and growth is on track, you could cut back to a few times a week or even a month for maintenance. If you find the oils too heavy, try mixing a few drops with your conditioner, or a shampoo for thinning hair. How long does it take to see results from hair growth oils? How long it takes for you to see a difference in your hair depends on a variety of factors, like diligence, hair oil type, and the condition of your hair when you begin your regimen. With consistent use, hair growth oils and serums may show subtle benefits in as little as six to eight weeks; however, full results typically require six to 12 months. You may also want to use a combination of products for thinning hair. 24 Best Prime Day Deals For Body Issues 2025

Amazon Prime Day is here! There are more deals than you could ever begin to look through on your that's where we come in. Up to 28% off a popular foot exfoliant with results that are equal parts disgusting and oh-so-satisfying. Within one to two weeks, the dead skin on your feet will literally start to peel off (gross!) and reveal the supremely soft skin that was hiding underneath (satisfying!). Check it out in action on TikTok !Promising review: "I was dubious at first, having seen this on TikTok. I can now confirm that it works as directed, and the results are amazing. 24 hours after I soaked my feet in the plastic bag, nothing happened. Then, 48 hours later, it started working. 72 hours, and it was in full working mode. Like other reviewers, I also suggest just letting the skin come off naturally. As someone who wears flip-flops often, I opted for socks and shoes during the shedding period to capture the skin, even when sleeping, because who wants chunks of dead skin in their bedsheets?! (it sounds so gross, but you'll understand once you do it) The shedding lasts for a few days. The results are incredible. Will use this again!" —VanessaPrice: $14.38 (originally $19.99; available in five scents) 50% off Philips Sonicare ~ExpertClean~ toothbrush (at its best price, and the first time it's been this price since *October 2022*) because if your manual toothbrush from the 1900s isn't giving you a deep cleaning to remove annoying food particles, this will! It comes with a charging case for traveling, and it even has a SENSOR THAT TELLS YOU IF YOU'RE BRUSHING TOO HARD. The technology we have!! Promising review: "I've been flying through a few of the cheapest sonicares for years, and after frustration, I caved and paid for this high-end one. It's really a treat to have a cadillac of a toothbrush! So many options. It's so nice to have a simple on and off button instead of one button controlling all the settings and taking forever to turn the brush off. This brush has one simple power button. A separate button to change the various modes to brush with and when the brush is on you can adjust power level. The brush comes with some really fancy brush heads, and they are a game changer. I might have to buy brand name brush heads after this. They're longer and curve to hit your teeth just right. You will instantly feel the difference after brushing with a sonicare, treat yourself to a higher end one. Your dental hygiene is worth it. You're worth it." —CharlottePrice: $99.96 (originally $199.95; available in three colors) — and shop all the Philips Sonicare deals here. Promising review: " Waterpik is amazing. I had 4mm and 5mm periodontal pockets from not flossing and also experienced some bone loss, I also had to get a very uncomfortable deep gum pocket cleaning. I literally hated flossing and would find myself avoiding to floss, even though I needed it very much. My gums were red, swollen, and inflamed because I was in the early stages of periodontal disease. I started using the Waterpik flosser and instantly liked this way of flossing much better! After using it consecutively each night for only five days my gums were noticeably better and felt much better. After two weeks of using the Waterpik flossing, coconut oil pulling, and mouthwash treatment, my gums have done a complete 180 and my mouth and gums are starting to heal properly! Also this flosser can be somewhat loud, but definitely gets the job done. Brought this with my own money and this is an honest testimonial/review. Reviewers *swear* by it for helping reduce stubborn blackheads and sebaceous filaments, too! As with all skincare products, if you're acne-prone YMMV, depending on the cause of your acne. Promising review: "I can't live without this cleansing oil; it's magic in a bottle. this is the only thing that's been able to clear the sebaceous filaments on my nose after consistent use daily!! it also helps the milia/filaments under my eyes! I wanted something small and nonaerosol (because travel), and I hate applying powder by hand and didn't want to bring an application brush. The application sponge is great, and the powder itself is also magic. I was a DAILY hair washer most of my teenage and adult life, but now I can go an entire WEEK with this stuff (and other mindful habits like night braids). I really love it."—BrePrice: $12.99+ (typically $18; available in six shades and in multipacks) 30% off Laneige's Neo Blurring Powder (the first time it's been on sale since it launched, and the LOWEST price it's ever been) to absorb that annoying oily layer that pops up outta nowhere. If I could recommend one product that is the key to glass skin, it would be this. It gives an immediate glow, and makes the skin look so supple and healthy. The texture wasn't off-puting for me, but it is a bit slimy and stringy when you first pump out the product. It absorbs pretty quickly and doesn't leave a sticky residue, and it is unscented. Don't hesitate, just buy" —SummerPromising review: "Where has this been all my life? I have been using this for maybe three days now, and putting it on in the morning and evening are the best parts of my day. The description of the scent as "pistachio, salted caramel, and vanilla" does not even begin to describe the incredibly decadent smell of this cream on my skin. My skin feels like velvet and I saw improvements in the texture after the first application. I will use it for the rest of my life. Do not ever discontinue this! I use this twice daily, and it's been a couple of weeks, and I definitely noticed my pores have shrunk. I even apply it to my neck and chest area. The product is soothing, gentle on my skin (I have extremely sensitive skin), has no strong smell, the cottons are soft, and did not cause any breakouts! A few dabs of this in the morning and at night will help get those sweaty dogs under control ASAP. Promising review: "Bought this for my boyfriend who is on his feet all day and was always complaining about how sweaty his feet got. He usually had to bring at least one extra pair of socks to change into during the day. He also had to buy shoes more often because of how they smell. It would be impossible for us to get out the smell. Since he began using this product, he can't believe how dry his feet stay ALL day. His socks don't even smell after a full day of work. In the words of this reviewer, "WHAT IN THE HAIR SORCERY IS GOING ON?!" The sorcery in question is the tiny fibers made with follicle-mimicking molecules that grab onto your hair strands and make it look like you took a magical overnight hair growth serum. Oh, and this is the lowest price we've seen the full size go for! To use, just sprinkle over the desired area and pat it in — you can also hit it with some hairspray for an extra secure hold. It'll wash out with your next shampoo. BuzzFeed Shopping editor Daniel Boan says: "I personally am OBSESSED with this stuff for covering the bald spot my thirties have so graciously gifted me. It is so simple to sprinkle into any thinning areas (yes, even by yourself! Though I typically hold up a hand mirror behind my head while looking in another mirror to get a better idea of where to aim the container). I've never had any issues with it fading or sweating off, though I do typically do a super light spritz of hairspray just in case."Promising reviews: "I have thinning hair due to a number of medical issues (thyroid, chemo), with the majority of bald/thin spots at the crown. The color match was perfect and it's very easy to apply to the bare spots. It sprinkles on easily and covers wonderfully. I pat it into place and finish with hairspray and I'm good to go." —mozart8"I have AA 4c hair that I recently cut super short. When it was long, my thin spots around the hairline were concealed. So, with my hair short, they were apparent and made me self-conscious. I purchased Boldify with just a little skepticism. I am so glad I purchased it. The application is easy and does not stain clothing. Be careful not to scratch your head throughout the day because you will see the dark coloring under your fingernails. It is a large bottle for the price. I will continue to buy and recommend." —Jza JzaPrice: $16.52+ (originally $22.95; available in four sizes and a two-pack, and in 14 colors). 36% off a Schick Silk dermaplaning razor that'll help you get rid of unwanted hair easily *and* painlessly. Many reviewers notice a reduction in hair growth in as little a 3-4 weeks. Note: This device is not designed to work on deeper skin tones. It also does not work for red, blonde, white, or gray hair. The set includes glasses that should be worn while operating the review: "So far, the hair growth has either slowed down to an incredible amount or completely stopped as of today. I went on a two-week trip mid-March where I couldn't use the device (overseas), and I barely had to shave any of these areas. For reference, before using this, I had to shave every day and would feel the hair growing back/stubble by evening on the same day, as my hair grows super fast. The hair that is growing back is much thinner and less noticeable, and I think with more use, it will almost completely disappear. I am SO happy I decided to try this out, and I think it was completely worth it. I am going to start using it on my legs soon." —SkaandarPrice: $198 (originally $249) — and shop all the Ulike deals here 30% off Biodance's TikTok-beloved Bio-Collagen Real Deep Masks you can wear overnight to improve dull skin with a serious boost of hydration, firming, and brightening — even reducing the appearance of pores and fine lines. Note: These typically go up to 32% off during Cyber Week and Prime Day, but they are SUPER popular, so this might be a good time to stock up before the word gets out 👀.Promising review: "Caught wind of this on TikTok and decided to give it a whirl. Honestly, it's pretty solid! It really makes my skin look tight and shiny, like I just came out of a professional facial. It's an overnight mask, so yeah, it does have to stay on for a while, which is a bit of a hassle, but the payoff? Totally worth it. My skin feels more elastic, and the pores look minimized, too. If you don't mind the long wear time, this mask could be a game changer for your skincare routine!" —NalaPrice: $13.30 for a set of four (typically $19; sometimes $15.20 at Amazon) — and shop all the Biodance deals here 30% off Mielle's (TikTok-famous) Rosemary Mint strengthening oil reviewers swear by to nourish their scalps, strengthening and promoting hair growth at the root. Took a chance on this because I was desperate, having already spent so much money on luxury and drugstore products with little results. I use this as a pre-treatment on scalp and ends before I wash hair. Scalp has improved dramatically. I feel like there's not as much breakage/fallout, and my ends are fuller. The shampoo is not runny, which is good because you have to leave it on the lathered hair for at least three minutes, and it doesn't get in my eyes during that time. The smell is not strong or medicinal; it is pleasant enough. It doesn't seem drying to my hair like some of the volumizing shampoos I've used. This product makes my hair feel so good and look so healthy. I will be buying this again!" —Rachel ElainePrice: $9.99 (typically $13.95; available in three scents).Psst — reviewers mention this is most effective when paired with a scalp massager to work it into their roots! 22% off a pack of cult-fave Mighty Patch pimple patches — these use hydrocolloid to absorb pus and oil from *inside* the zit, protecting skin from further damage and reinfection so you don't have to hot compress, pop, ooze, scar, and repeat. I am 23 years old, and I have struggled with stress for years, and it has caused cystic acne. I have tried so many different products, and I have pretty sensitive skin. These patches have relieved so much pressure in my acne and have cleared them up beyond words! Seriously, these honestly work!! I will be a loyal customer for sure. I cannot thank this product enough. I just got over my period and had awful cystic acne. Usually, my acne would be in pain for a whole week after my period... I used these patches right away when I felt pressure in a spot on my face. My period is now over, and my face is completely cured and healing with minor scars!! The amount of disgusting gunk that came out of my face was appalling and I instantly gagged. However, my nose has never felt so clean and smooth for consecutive days. If you have oily skin I definitely recommend washing your face first and apply it to the nose before moisturizing the rest of your face. If I could give this 10 stars I would. Everyone is getting this for Christmas….maybe because it's not cheap." —Justina"Saw it on TikTok and already use their acne patches, so I was sold. Works just as well. I'd highly recommend them!" —Sara Fox Price: $13.24 for 10 patches (originally $16.55) 25% off a portable neck fan because, let's be honest, does anyone enjoy having a sweaty face or neck? I was tired of always having to move my portable fan and charger pack wherever I went. I use this all. the. time! I usually do the lowest setting, and it usually lasts the whole day. I'm super grumpy when I'm hot so this fan probably saved our marriage. Bonus! When it's running I can't hear my son talk about Roblox or Minecraft. He thinks I'm listening but I can't hear him. 🤷‍♀️"—Randall McCormickPrice: $23.98 (originally $31.99)

Solid Biosciences Announces FDA IND and Health Canada CTA Approval for First-in-Class Cardiac Gene Therapy to Treat Catecholaminergic Polymorphic Ventricular Tachycardia (CPVT)

- SGT-501 is a novel gene therapy for rare, life-threatening, genetic arrhythmogenic disease with no approved therapies - - SGT-501 has received Orphan Drug Designation and Rare Pediatric Disease Designation from the FDA - - Solid expects to initiate Phase 1b clinical trial of SGT-501 in the fourth quarter of 2025 - - Expands Solid's clinical pipeline to include first cardiac indication with urgent unmet medical need - CHARLESTOWN, Mass., July 08, 2025 (GLOBE NEWSWIRE) -- Solid Biosciences Inc. (Nasdaq: SLDB) (the 'Company' or 'Solid'), a life sciences company developing precision genetic medicines for neuromuscular and cardiac diseases, today announced approval of its Investigational New Drug (IND) application by the U.S. Food and Drug Administration (FDA) and clinical trial application (CTA) by Health Canada for SGT-501, a novel gene therapy approach for the treatment of catecholaminergic polymorphic ventricular tachycardia (CPVT), a highly malignant, arrhythmogenic channelopathy caused by genetic mutations that impact the ryanodine receptor (RYR2) in cardiac muscle. The Company expects to initiate a Phase 1b clinical trial to evaluate the safety, tolerability and efficacy of SGT-501 in the fourth quarter of 2025. Gabriel Brooks, M.D., Chief Medical Officer of Solid, said: 'Despite being identified nearly 50 years ago, CPVT still lacks FDA-approved therapies – this announcement reflects a critical development in the treatment of this underserved, often fatal, cardiac disease. SGT-501 offers a precision genetic approach targeting the underlying pathophysiology of the disease: abnormal calcium releases from the sarcoplasmic reticulum in an otherwise structurally sound heart. We believe SGT-501 has the unique potential to provide durable protection and may be capable of liberating patients from the ever-present threat of lethal arrhythmias and life-limiting prohibitions on exercise. Solid is proud to launch this program to help address this clear unmet need, further expanding our pipeline of differentiated and thoughtfully designed genetic medicines.' Silvia Priori, M.D., Ph.D., Professor of Cardiology at the University of Pavia and Director of the Molecular Cardiology Unit at the IRCCS Maugeri in Pavia, Italy, said: 'After decades during which we, the clinical community, have been limited in our ability to treat people living with CPVT, I have long hoped for the day when a genetic-modifying therapy becomes available. We are proud to partner with Solid who advanced fundamental work from our labs at Maugeri, which demonstrated the principle that calsequestrin overexpression can have a therapeutic impact on multiple forms of CPVT in both cellular and mouse models of disease. This work was further developed into a compelling IND safety and efficacy package through close collaboration with, and studies conducted by, the Solid team. I look forward to clinical updates for SGT-501 and seeing how this potential medicine may benefit people living with CPVT who are in critical need of disease-specific medicines.' About SGT-501SGT-501 is an AAV-based gene therapy candidate designed to deliver a functional, full length, codon-optimized copy of the human cardiac calsequestrin (CASQ2) gene to heart muscle cells. In the context of RYR2 variants, increasing CASQ2 protein levels enhances buffering of free calcium in the sarcoplasmic reticulum, stabilizing the RYR2, which results in reduced diastolic calcium leak into the cytosol. Stabilization of the RYR2 in its closed conformation supports the maintenance of normal cardiac rhythm with the potential to protect against ventricular tachycardia. SGT-501 received Orphan Drug Designation and Rare Pediatric Disease Designation from the U.S. FDA and has potential as a first-in-class therapy to correct the underlying RYR2 instability and calcium dysregulation causes of CPVT. The AAV-CASQ2 gene therapy approach utilized by SGT-501 was pioneered by Dr. Silvia Priori and the IRCCS ICS Maugeri lab in Pavia, Italy. As a globally recognized leader in inherited arrhythmias, Dr. Priori has long been at the forefront of translational cardiac research. Solid Biosciences in-licensed this innovative therapeutic in 2023 and subsequently advanced it into IND-enabling preclinical development, marking an important step toward bringing this potential treatment to individuals living with CPVT. About Catecholaminergic Polymorphic Ventricular Tachycardia (CPVT)Catecholaminergic polymorphic ventricular tachycardia (CPVT) is a heart condition marked by abnormal rhythms; specifically, an abnormally fast heartbeat (ventricular tachycardia), which can be triggered by adrenergic stimulation such as physical activity or emotional stress. These arrhythmias can cause unexplained fainting, seizures, cardiac arrest and sudden death. CPVT is estimated to affect roughly 1 in 10,000 individuals globally, although it is often misdiagnosed, and is typically identified in children and young adults. It is mainly caused by mutations in the RYR2 and CASQ2 genes, which disrupt calcium regulation in heart muscle cells, impairing proper heart contraction and relaxation. There are currently no treatments that address the underlying mechanisms of CPVT. To learn more about CPVT and SGT-501, watch a brief CPVT expert and patient video by clicking here.
