Study on Mice Suggests Surprising Link Between Nose-Picking And Alzheimer's

A study published in 2022 revealed a tenuous but plausible link between picking your nose and increasing the risk of developing dementia.
In cases where picking at your nose damages internal tissues, critical species of bacteria have a clearer path to the brain, which responds to their presence in ways that resemble signs of Alzheimer's disease.
There are plenty of caveats here, not least that so far the supporting research is based on mice rather than humans, but the findings are definitely worth further investigation – and could improve our understanding of how Alzheimer's gets started, which remains something of a mystery.
A team of researchers led by scientists from Griffith University in Australia ran tests with a bacterium called Chlamydia pneumoniae, which can infect humans and cause pneumonia.
The bacteria has also been discovered in the majority of human brains affected by late-onset dementia.
It was demonstrated that in mice, the bacteria could travel up the olfactory nerve (joining the nasal cavity and the brain). What's more, when there was damage to the nasal epithelium (the thin tissue along the roof of the nasal cavity), nerve infections got worse.
This led to the mouse brains depositing more of the amyloid-beta protein – a protein which is released in response to infections. Plaques (or clumps) of this protein are also found in significant concentrations in people with Alzheimer's disease.
"We're the first to show that Chlamydia pneumoniae can go directly up the nose and into the brain where it can set off pathologies that look like Alzheimer's disease," said neuroscientist James St John from Griffith University in Australia back in October 2022, when the study was released.
"We saw this happen in a mouse model, and the evidence is potentially scary for humans as well."
The scientists were surprised by the speed at which C. pneumoniae took hold in the central nervous system of the mice, with infection happening within 24 to 72 hours. It's thought that bacteria and viruses see the nose as a quick route to the brain.
While it's not certain that the effects will be the same in humans, or even that amyloid-beta plaques are a cause of Alzheimer's, it's nevertheless important to follow up promising leads in the fight to understand this common neurodegenerative condition.
"We need to do this study in humans and confirm whether the same pathway operates in the same way," said St John.
"It's research that has been proposed by many people, but not yet completed. What we do know is that these same bacteria are present in humans, but we haven't worked out how they get there."
Nose picking isn't exactly a rare thing. In fact, it's possible as many as 9 out of 10 people do it… not to mention a bunch of other species (some a little more adept than others). While the benefits aren't clear, studies like this one should give us pause before picking.
Future studies into the same processes in humans are planned – but until then, St John and his colleagues suggest that picking your nose and plucking your nose hair are "not a good idea" because of the potential damage it does to protective nose tissue.
One outstanding question that the team will be looking to answer is whether or not the increased amyloid-beta protein deposits are a natural, healthy immune response that can be reversed when the infection is fought off.
Alzheimer's is an incredibly complicated disease, as is clear from the sheer number of studies into it and the many different angles scientists are taking in trying to understand it – but each piece of research brings us a little bit closer to finding a way to stop it.
"Once you get over 65 years old, your risk factor goes right up, but we're looking at other causes as well, because it's not just age – it is environmental exposure as well," said St John.
"And we think that bacteria and viruses are critical."

Scientists Find 2 Existing Drugs Can Reverse Alzheimer's Brain Damage in Mice

In efforts to beat Alzheimer's disease, researchers are looking at existing drugs that could tackle the condition, and a new study identifies two promising candidates that are currently used to treat cancer. Already approved by regulators in the US – meaning potential clinical trials for Alzheimer's could start sooner – the drugs are letrozole (usually used to treat breast cancer) and irinotecan (usually used to treat colon and lung cancer). Researchers from the University of California, San Francisco (UCSF) and Gladstone Institutes started by looking at how Alzheimer's altered gene expression in the brain. They then consulted a medical database called the Connectivity Map to look for drugs that reversed these gene expression changes, and cross-referenced records of patients who had taken these drugs as part of cancer treatments and their likelihood of developing Alzheimer's. Intriguingly, the drugs seemed to have reduced their risk. Related: "Alzheimer's disease comes with complex changes to the brain, which has made it tough to study and treat, but our computational tools opened up the possibility of tackling the complexity directly," says computational biologist Marina Sirota, from UCSF. "We're excited that our computational approach led us to a potential combination therapy for Alzheimer's based on existing FDA-approved medications." Having picked out letrozole and irinotecan as the best candidates, the researchers tested them in mouse models of Alzheimer's. When used in tandem, the drugs were shown to reverse some of the brain changes brought on by the disease. The harmful clumps of tau protein that build up in brains affected by Alzheimer's were reduced significantly, and the mice showed improvements in learning and memory tasks – two brain capabilities often impaired by Alzheimer's. By combining the two drugs together, the researchers were able to target different types of brain cells affected by the disease. Letorozole seemed to counter Alzheimer's in neurons, while irinotecan worked in glia. "Alzheimer's is likely the result of numerous alterations in many genes and proteins that, together, disrupt brain health," says neuroscientist Yadong Huang, from UCSF and Gladstone. "This makes it very challenging for drug development – which traditionally produces one drug for a single gene or protein that drives disease." It's a promising start, but there's more work to be done: obviously the drugs have only been directly tested in mice so far, and these medications also come with side effects attached. They need to be reconsidered if the drugs are going to be repurposed for a different disease than what they were originally approved for. One of the next steps should be clinical trials for people with Alzheimer's disease. According to the researchers, this approach could lead to more personalized and effective treatments, based on how gene expression has been altered in each case. It's estimated that more than 55 million people have Alzheimer's today, and as the world's population ages, that's expected to more than double in the next 25 years. Finding ways to prevent the disease and even reverse symptoms would have a huge impact on global health. "If completely independent data sources, such as single-cell expression data and clinical records, guide us to the same pathways and the same drugs, and then resolve Alzheimer's in a genetic model, then maybe we're on to something," says Sirota. "We're hopeful this can be swiftly translated into a real solution for millions of patients with Alzheimer's." The research has been published in Cell. These 4 Distinct Patterns May Signal Alzheimer's According to Science

There is no cure for Alzheimer's disease yet, but detecting it early can make a big difference. It gives scientists a better chance to study it in depth and gives patients and families more time to plan and get support. Now, US researchers have uncovered four telltale medical sequences that appear to predict the onset of the progressive neurological disorder. In the large new study, scientists analyzed health records from 24,473 people diagnosed with Alzheimer's, searching for patterns that typically led up to their diagnosis – and crucially, how different factors combined in sequence. "We found that multi-step trajectories can indicate greater risk factors for Alzheimer's disease than single conditions," says Mingzhou Fu, a bioinformatician at the University of California, Los Angeles (UCLA). "Understanding these pathways could fundamentally change how we approach early detection and prevention." Related: Four "trajectory clusters" were identified by the team, which are essentially different routes to Alzheimer's, like step-by-step directions towards a destination on a map. Analysis of an independent dataset from across the US found people who follow one of these trajectories tend to have a significantly higher risk of Alzheimer's. The four clusters were mental health (psychiatric conditions), encephalopathy (brain dysfunctions that gradually get worse), mild cognitive impairment (declining mental dexterity), and vascular disease (heart and blood conditions). The researchers used an algorithmic approach called dynamic time warping to standardize the duration and sequence of health issues across these thousands of database records, and find patterns that matched up between patients. In the mental health cluster, for example, anxiety often came first, followed by depression, which often eventually led to Alzheimer's. In the vascular cluster, conditions like hypertension and joint disorders were common starting points on the road to Alzheimer's. Across the four clusters, thousands of different individual trajectories were identified, varying in terms of their progression speeds and associated Alzheimer's risk level. It shows just how complex the road to Alzheimer's can be. "By unveiling distinct and interconnected routes to Alzheimer's disease, this approach offers insights that may improve risk assessment, timely diagnosis, and targeted interventions," write the researchers in their published paper. To test their findings, the team ran their model on a separate set of health records from 8,512 people. The pathways identified by the study were far more likely to be present in those with Alzheimer's, adding weight to the results. Of course, understanding more about how Alzheimer's progresses in the body could help us stop it: there may be blocks that can be put in place along the way that prevent the disease from fully developing, or at least reduce the risk. As always, there's plenty more work to do here. The researchers want to study broader groups of people with and without Alzheimer's to further validate their findings, and expand the work to more types of dementia. While these four clusters don't imply direct cause and effect or guarantee someone will develop Alzheimer's, they're likely to be important in future assessments of patients – and in figuring out how to stop this devastating disease. "Recognizing these sequential patterns rather than focusing on diagnoses in isolation may help clinicians improve Alzheimer's disease diagnosis," says neurologist Timothy Chang, from UCLA. It's official: Older adults worried about cognitive decline can stay sharper for longer by exercising both their bodies and their brains and eating healthier. That's according to initial results released Monday from a rigorous US study of lifestyle changes in seniors at risk of developing dementia. People following a combination of healthier habits slowed typical age-related cognitive decline — achieving scores on brain tests as if they were a year or two younger, researchers reported in the medical journal JAMA. It's not too late to get started — study participants were in their 60s and 70s — and it doesn't require becoming a pickleball champ or swearing off ice cream. 'It was the first time I felt like I was doing something proactive to protect my brain,' said Phyllis Jones, 66, from the US state of Illinois. She joined the study after caring for her mother with dementia and struggling with her own health problems. It's too soon to know if stalling age-related decline also could reduce the risk of later Alzheimer's or other forms of dementia. But Jones and other study participants underwent brain scans and blood tests that researchers now are analysing for clues — such as whether people also saw a reduction in Alzheimer's-related protein buildup. Related New dementia patients can expect to live fewer than five years after diagnosis, major analysis finds 'We're all on a cognitive ageing clock and anything we can do to slow that clock down, to me, that is a significant benefit,' said Laura Baker of Wake Forest University School of Medicine, who led the study. Doctors have long encouraged physical activity and a healthy diet for brain fitness. Those steps fight high blood pressure and cholesterol, heart disease, and diabetes, factors that increase the risk of dementia. But until now the strongest evidence that specific lifestyle changes later in life could improve how people perform on brain tests came from a study in Finland. How the study worked Half of participants were randomly assigned to group classes for exercise and dietary changes plus brain-challenging homework — with peer support and coaches tracking their progress. They did a half-hour of moderately intense exercise four times a week — plus twice a week, they added 10 to 15 minutes of stretching and 15 to 20 minutes of resistance training. They followed the 'MIND diet' that stresses lots of leafy greens and berries plus whole grains, poultry, and fish. Nothing is banned, but it urges limiting red meat, fried or fast food, and sweets, and substituting olive oil for butter and margarine. Other study participants, the control group, received brain-healthy advice and minimal coaching — they chose what steps to follow. Both improved but the group with more support fared significantly better. Combining social engagement with exercise and dietary steps may be key, said Jessica Langbaum of the Banner Alzheimer's Institute, who wasn't involved with the study. 'Americans want to have that one easy thing — 'If I just eat my blueberries,'' Langbaum said. 'There is no one magic bullet. It is a whole lifestyle". How to exercise your body and mind on your own Moderately intense physical activity means raising your heart rate and panting a bit yet still being able to talk, said Wake Forest's Baker. Pick something safe for your physical capability and start slowly, just 10 minutes at a time until you can handle more, she cautioned. Likewise there are many options for brain exercise, Baker said — puzzles, joining a book club, learning an instrument or a new language. One challenge: How to keep up the good work Researchers will track study participants' health for four more years, and the Alzheimer's Association is preparing to translate the findings into local community programmes. Will people with stick with their new habits? Jones lost 30 pounds, saw her heart health improve and feels sharper, especially when multitasking. But she hadn't realised her diet slipped when study coaching ended until a checkup spotted rising blood sugar. Now she and an 81-year-old friend from the study are helping keep each other on track. The lifestyle change 'did not just affect me physically, it also affected me mentally and emotionally. It brought me to a much better place,' Jones said.
