Stem Cell Treatment to Reverse Hearing Loss Kicking Off in Human Patients
Conducted by researchers from the University of Sheffield who formed their own spin-out company, the treatment targets sensorineural hearing loss, which is caused by physical damage to the tiny structures of the inner ear.
In a nutshell, the treatment, dubbed Rincell-1, is intended to regrow damaged nerves in the cochlea and allow them to start sending signals to the brain again.
"Our research into Rincell-1 has consistently shown its ability to target and restore the delicate neural structures of the inner ear," Marcelo Rivolta from the University of Sheffield, chief scientific officer at the spin-out biotech company Rinri Therapeutics, said in a statement about the work.
More than a billion people worldwide are affected by some form of hearing loss, according to the WHO. Existing treatments, like cochlear implants, don't fully restore a patient's hearing, and require that the cochlea — the center piece of the inner ear that picks up sound and turns it into electrical signals — isn't significantly damaged to begin with.
Key to the cochlea's function are the hair cells that line its surface, which are responsible for detecting sound. If these are severely damaged, it's game over: the hair cells are incapable of dividing to form new ones, meaning that they don't regenerate. This is called sensorineural hearing loss, and it's why your hearing inevitably worsens with age.
Attempts to regrow these receptors have long been the white whale of modern medical science, and the researchers hope they've found it in Rincell-1. The treatment uses embryonic stem cells designed to grow into auditory neuron cells, which form the wiring that connects the hair cells to the brain stem. These are administered into the cochlea during the surgery to emplace the cochlear implants. Once on site, the stem cells form auditory neurons that help reconnect the out-of-commission hair cells so they can send signals again.
"We are taking the approach of transplanting in cells that can become functional mature cells and restore the cytoarchitecture of the inner ear, and therefore, restore hearing," Simon Chandler, CEO of Rinri Therapeutics, told Labiotech on an episode of its "Beyond Biotech" podcast in June.
The randomized trial will be conducted in the UK and will involve 20 patients who will undergo cochlear implant surgery. Half of the patients will have severe-to-profound age-related hearing loss, known as presbycusis, and the remainder will have what's known as auditory neuropathy spectrum disorder (ANSD), which is when the inner ear can detect sound but is unable to send the signals to the brain. Patients within each group will be randomly assigned to either receive a single dose of Rincell-1, or receive no dose and rely on the cochlear implant alone.
On the podcast, Chandler said that the Rincell-1 treatment can be used alongside cochlear implants, but didn't rule out the possibility that it would be used on its own, too. Needless to say, it'd be a game-changer if it turns out to be effective in either scenario.

I'd just had a baby and life seemed perfect – then I got the worst possible news
Having a baby is supposed to be one of the most joyous occasions imaginable. But for Beth Jenkins it brought a world of pain she would never have expected. While 40-year-old Beth had the delight of her second daughter Robyn she believes her pregnancy caused her to have breast cancer, which was not initially spotted by a doctor who told her she was "too young" to have it. Knowing that cancer can happen to anyone of any age, and sticking with her intuition when she found a lump in her breast, Beth fought for answers. She went on to give birth to her daughter Robyn in 2023. As she was set to return to work the following year she noticed the lump in her breast had grown and become painful after her daughter had kicked her in the chest. After being examined by a doctor she was told it was just an infection and that she was "too young to have breast cancer". Beth felt this diagnosis could be wrong and more needed to be done to determine what the lump was. She said: "They were quite dismissive and passive. They began asking me questions about my postnatal depression but it was a physical lump that he felt so I don't know what that would have had to do with it." A few weeks later at a check-up for her daughter Beth expressed her concern to another doctor who told her she should have been immediately referred to the hospital for a scan. Contacting her own doctor once again Beth was referred to a breast clinic that was able to see her within two weeks. Taking her dad along she was hoping for answers that would put her mind at rest. But after an examination was followed by an ultrasound and a mammogram a consultant told the mum-of-two she had breast cancer. A biopsy followed and two weeks later Beth and her husband Lee spoke with the consultant to confirm the type of breast cancer and discuss a treatment plan. Beth was told that the cancer was ER-positive, PR-positive, and HER2-negative, meaning that it was brought on by her hormones that had changed during pregnancy. "I didn't have a clue that this could happen – that's what's scary," she said. Her biopsy also confirmed that the cancer had spread. "If I hadn't have gone back to the doctors and I hadn't listened to my body I don't know where I'd be now," she added. "It had already spread to my lymph nodes. If I had listened when I was told it was an infection I would be looking at a totally different story now." In December 2024 the tumour in Beth's breast was 5cm which meant that she had to have a full breast mastectomy with reconstruction as well as having lymph nodes removed from her arm. Although she was still recovering from surgery she had on December 18 Beth managed to cook a full Christmas dinner for her family to keep everything "as normal as possible" for her nine-year-old daughter Fallon. After starting extensive chemotherapy in February this year Beth currently has two sessions left before she will need radiotherapy, hormonotherapy, and bone therapy. Due to the cancer being caused by hormonal changes Beth will also be put into early menopause as there is a high risk of the cancer returning when her hormones change. Beth has said one of the hardest experiences throughout the process was having to tell her daughter that she had cancer, which she did as hair began to fall out during her treatment. Beth's daughter Fallon had already witnessed the effects of cancer as her grandfather, Beth's father, has had cancer seven times. Beth said: "With my dad having cancer seven times she's seen that cancer doesn't mean you're going to die. If anything it helped the situation to say: 'Look – he's had cancer and he's still here'." Throughout her diagnosis Beth has remained brave and shown her two daughters that while you can't control whether you have cancer or not you can control your mindset. She said: "The word cancer is scary enough. She's being pulled out of classes in school because she gets upset and she is having support in school. "She is really sensitive and she does break down crying but I tell her that everything will be all right. "It is hard."
