
UK scientists to synthesise human genome to learn more about how DNA works
Researchers are embarking on an ambitious project to construct human genetic material from scratch to learn more about how DNA works and pave the way for the next generation of medical therapies.
Scientists on the Synthetic Human Genome (SynHG) project will spend the next five years developing the tools and knowhow to build long sections of human genetic code in the lab. These will be inserted into living cells to understand how the code operates.
Armed with the insights, scientists hope to devise radical new therapies for the treatment of diseases. Among the possibilities are living cells that are resistant to immune attack or particular viruses, which could be transplanted into patients with autoimmune diseases or with liver damage from chronic viral infections.
'The information gained from synthesising human genomes may be directly useful in generating treatments for almost any disease,' said Prof Jason Chin, who is leading the project at the MRC's Laboratory of Molecular Biology (LMB) in Cambridge.
Scientists have been able to read DNA for decades. The first draft of the human genome was announced 25 years ago, a feat that set the stage for the ongoing genetics revolution. But while the technology for reading genomes has progressed rapidly, writing them has proved more difficult.
For the SynHG project, researchers will start by making sections of a human chromosome and testing them in human skin cells. The project involves teams from the universities of Cambridge, Kent, Manchester, Oxford and Imperial College London.
Chin's team recently synthesised the complete genome of the E coli bacterium. But while the bug's genome carries about 4.5m base pairs, represented by the letters G, T, C and A, the human genome holds more than 3bn base pairs.
'If you think about the human genome, it's more than just a set of genes on a string,' said Dr Julian Sale, a group leader at the LMB. 'There's an awful lot of the genome, sometimes called the dark matter of the genome, that we don't know what it does. The idea is that if you can build genomes successfully, you can fully understand them.'
Embedded in the project is a parallel research effort into the social and ethical issues that arise from making genomes in the laboratory, led by Prof Joy Zhang at the University of Kent. 'We're a little way off having anything tangible that can be used as a therapy, but this is the time to start the discussion on what we want to see and what we don't want to see,' Sale said.
Prof Iain Brassington, who studies the ethics of genetic technology at the University of Manchester, welcomed the project. While real-world applications are some way off, he said the work could lead to synthetic versions of mitochondria, the tiny battery-like structures that power biological cells. These could be used to prevent women who carry mitochondrial diseases from passing them on to their children. 'The would-be mother would still need to undergo egg harvesting and IVF, but we'd no longer need a donor, so we'd halve the number of women burdened by the procedure,' he said.
But the technology did pose problems, Brassington added. It might be possible to make bacteria that – thanks to their synthetic genomes – can digest petrochemicals, which could be useful for breaking down plastic waste and clearing up oil spills but would need to be handled extremely carefully. 'Such bugs getting into the environment could be catastrophic,' he said.
The spectre of designer babies is another valid concern. Parents could potentially use the technology to shape their children before birth, raising questions over how much control they should have. In another 'slightly wild' idea, Brassington said celebrities might even start 'licensing' parts of their genome to allow people to copy their genes.
'There might also be situations in which a man discovers that he is the 'genetic' parent of a child of whom he knew nothing,' Brassington added. 'This could already happen, of course, but he would currently know how it happened. SynDNA enables us to imagine a situation in which a replica of someone's DNA could be used to father a child with no cellular contribution from him at all.'
However, he said: 'Generally, I think the concerns about designer babies all rely on some fairly far-fetched technology. I'm not going to lose too much sleep over them.'

Variety has learned from insiders that the new iteration of 007 would be under 30 and the three actors could be fighting it out for the role. The French-Canadian film-maker, now working on the third Dune movie, reportedly beat out Conclave's Edward Berger, Westworld's Jonathan Nolan, Paddington's Paul King and Shaun of the Dead's Edgar Wright for the role. 'I intend to honour the tradition and open the path for many new missions to come,' Villeneuve said in a statement. 'This is a massive responsibility, but also, incredibly exciting for me and a huge honour.' Early rumours had suggested the Gravity director Alfonso Cuarón would step up, having worked with Bond's new producer David Heyman before, but he removed himself from the race. Cuarón is set to work on the darkly funny drama Jane with Charlize Theron instead. Holland, best known for playing Spider-Man, has also starred in the video game hit Uncharted and Apple series The Crowded Room. He'll next be seen in Christopher Nolan's The Odyssey before returning for the fourth Spider-Man movie. The 29-year-old actor once tried to pitch a 007 spin-off to Sony. 'I had a meeting after or during Spider-Man 2 with Sony to pitch this idea of a young Bond film that I'd come up with,' he said in 2022. 'It was the origin story of James Bond. It didn't really make sense. It didn't work. It was the dream of a young kid, and I don't think the Bond estate were particularly interested.' Dickinson, known for Triangle of Sadness and Babygirl, recently received rave reviews for his directorial debut Urchin at the Cannes film festival. 'I mean, listen, man, you'd be a fool to not entertain that role,' he said when asked about playing Bond in 2023. Wes Streeting is right to listen to bereaved parents – and I am one of them
How do you parent a baby who has died? This is a question my partner and I were left with when our son, Sasha, was stillborn in January 2022. We had all this overflowing love and care in our hearts, and no living baby to give it to. People don't like to think about baby loss. It's simply too sad and shocking for most. So, for us, keeping Sasha's name alive and trying to speak the truth of his life and death became one of the ways we parented him. Being unheard is one of the most common experiences that unites bereaved families. It is not only that many of us fear stigma or hurtful reactions when we speak about our babies. Many parents, especially those from low-income or ethnic minority families, will have had their concerns during pregnancy ignored or not taken seriously by healthcare professionals. When the tragedy of a baby's death occurs, it is often the case that our complaints are met with denial or obfuscation by the hospital. Many bereaved parents, particularly those who feel or know that their baby's death could have been prevented, eventually come to expect that we will not be listened to, that our warnings or suggestions about maternity care in the UK will remain unheeded. Now, I can say for the first time since my boy died, I know what it feels like to be listened to. When Wes Streeting invited bereaved families to speak with him about their losses and what needs to improve in UK maternity services, many expected that it would not lead to any meaningful change. And this nearly did happen. But after presenting an overview of suggested actions to improve maternity services to bereaved parents – including those whose losses occurred at scandal-hit hospitals such as Nottingham, Shrewsbury and Telford, Leeds and Sussex – something did change. The health secretary openly acknowledged that the conventional approach to policy was not working. He said that he would work more closely with bereaved parents and head a task force that would include bereaved parents, promising to produce this policy together. Witnessing the words of bereaved parents genuinely convince the health secretary and change the course of action felt like a miracle. Of course, it shouldn't have. This should be how most decisions in our country are made – with the direct involvement of those closest to the issue. Not only did I feel lighter, but, for the first time since my son's death, I felt hope that things might actually change. Wes Streeting has realised something that many policymakers and NHS leaders before him have been unable to see. Bereaved parents are key to improving maternity care. We are not only experts by experience, having witnessed firsthand how things go wrong in maternity care. Whether by advocating for change, pursuing legal justice or following complaints procedures, we also amass a depth of knowledge of the wider healthcare system, from both a user and a technical experience perspective. This is only the start of a much larger and longer process, but this new beginning is on the right track. I hope that policymakers continue to listen to us, to seriously consider our suggestions and learn from our losses, even though it is a more challenging process than traditional top-down decision making. I hope that they realise that we are their biggest and most trustworthy asset for improving maternity care. We have nothing to personally gain, no reputations to protect, and we have already lost that which is most precious to us. We simply don't want future parents and babies to go through what we did. We want a future where parents can be confident they will receive safe care and be listened to when this goes wrong.
