
In US, first baby treated with custom gene-editing, sparking hope for rare illnesses
The wee pioneer is KJ Muldoon, now a nine-and-a-half-month-old boy with chubby cheeks and big blue eyes.
Shortly after birth, he was diagnosed with a rare and serious condition called CPS1 deficiency.
It is caused by a mutation in a gene that produces an enzyme key to liver function, and prevents people with it from eliminating certain kinds of toxic waste produced by their metabolism.
'You Google 'CPS1 deficiency' and it's either fatality rate or liver transplant,' the baby's mother, Nicole Muldoon, says in a video released by Children's Hospital of Philadelphia, where the baby was treated.
With the prognosis grim, doctors suggested something that had never been done before: a personalised treatment to fix the baby's genome using what amounts to a pair of molecular scissors — the technique called Crispr-Cas9, which earned its creators the Nobel prize for chemistry in 2020.
The boy's father said he and his wife faced an impossible decision.
'Our child is sick. We either have to get a liver transplant or give him this medicine that's never been given to anybody before, right?' said Kyle Muldoon.
In the end, they agreed to have the child treated with an infusion created just for him to fix his genetic mutation — incorrect DNA letters in the several billion that make up the human genome.
'The drug is really designed only for KJ, so the genetic variants that he has are specific to him. It's personalised medicine,' said Rebecca Ahrens-Nicklas, a member of the medical team who specialises in paediatric genetics.
Once the tailor-made infusion reaches the liver, the molecular scissors contained in it penetrates cells and goes to work editing the boy's flawed gene.
The results were promising for other people with genetic conditions, said the medical team, which published their study Thursday in the New England Journal of Medicine.
KJ can now follow a diet richer in proteins — his condition prohibited such before — and does not need as much medicine as he used to.
But he will need to follow-up long term to monitor the safety and efficacy of the treatment, the team said.
Ahrens-Nicklas said she hoped this achievement will allow the boy to get by with little or no medication some day.
'We hope he is the first of many to benefit from a methodology that can be scaled to fit an individual patient's needs,' the doctor said. — AFP

LOS BANOS (US), July 6 — Oblivious to the punishing midday heat, a wheeled robot powered by the sun and infused with artificial intelligence carefully combs a cotton field in California, plucking out weeds. As farms across the United States face a shortage of labourers and weeds grow resistant to herbicides, startup Aigen says its robotic solution — named Element — can save farmers money, help the environment and keep harmful chemicals out of food. 'I really believe this is the biggest thing we can do to improve human health,' co-founder and chief technology officer Richard Wurden told AFP, as robots made their way through crops at Bowles Farm in the town of Los Banos. 'Everybody's eating food sprayed with chemicals.' Wurden, a mechanical engineer who spent five years at Tesla, went to work on the robot after relatives who farm in Minnesota told him weeding was a costly bane. Weeds are becoming immune to herbicides, but a shortage of labourers often leaves chemicals as the only viable option, according to Wurden. 'No farmer that we've ever talked to said 'I'm in love with chemicals',' added Aigen co-founder and chief executive Kenny Lee, whose background is in software. 'They use it because it's a tool — we're trying to create an alternative.' Aigen's vision is for workers who once toiled in the heat to be 'upskilled' to monitor and troubleshoot robots. Study says over 14 million people could die from US foreign aid cuts

(AFP pic) PARIS : More than 14 million of the world's most vulnerable people, a third of them small children, could die by 2030 because of the Trump administration's dismantling of US foreign aid, research projected today. The study in the prestigious medical journal The Lancet was published as world and business leaders met at a United Nations conference in Spain to try to bolster the reeling aid sector. The US Agency for International Development (USAID) had provided over 40% of global humanitarian funding until Donald Trump returned to the White House in January. Two weeks later, Trump's then-close adviser – and the world's richest man – Elon Musk boasted of having put the agency 'through the woodchipper'. The funding cuts 'risk abruptly halting – and even reversing – two decades of progress in health among vulnerable populations', warned study co-author Davide Rasella, a researcher at the Barcelona Institute for Global Health (ISGlobal). 'For many low and middle-income countries, the resulting shock would be comparable in scale to a global pandemic or a major armed conflict,' he said in a statement. Looking back over data from 133 nations, the international team of researchers estimated that USAID funding had prevented 91.8 million deaths in developing countries between 2001 and 2021. That is more than the estimated number of deaths during World War II – history's deadliest conflict. Children already dying The researchers also used modelling to project how funding being slashed by 83% – the figure announced by the US government earlier this year – could affect death rates. The cuts could lead to more than 14 million avoidable deaths by 2030, the projections found. That number included over 4.5 million children under the age of five – or around 700,000 child deaths a year. For comparison, around 10 million soldiers are estimated to have been killed during World War I. USAID funding was found to be particularly effective at staving off preventable deaths from disease. There were 65% fewer deaths from HIV/AIDS in countries receiving a high level of support compared to those with little or no USAID funding, the study found. Deaths from malaria and neglected tropical diseases were similarly cut in half. In South Sudan, people are already dying as a result of the US cuts, said Denish Ogen Rwot of the NGO Action Against Hunger, which has slashed its staffing in the country from 300 to 86. 'Already we are having children die,' he told AFP. A recently updated tracker run by disease modeller Brooke Nichols at Boston University estimates that nearly 108,000 adults and more than 224,000 children have already died as a result of the US aid cuts. That works out to 88 deaths every hour, according to the tracker. More deaths from Europe, UK cuts After USAID was gutted, several other major donors, including France, Germany and the UK, followed suit in announcing plans to slash their foreign aid budgets. These aid reductions, particularly in the European Union, could lead to 'even more additional deaths in the coming years', study co-author Caterina Monti of ISGlobal said. But the grim projections are based on the current amount of pledged aid, so could rapidly come down if the situation changes, the researchers emphasised. Dozens of world leaders – though no one from the US – attended an aid conference in the Spanish city of Seville this week. A common declaration was adopted reaffirming previous goals but it is not legally binding. This year's aid cuts spearheaded by the US could also nearly double the number of people forced to flee their homes this year, the Danish Refugee Council warned today. Before its funding was slashed, USAID represented 0.3% of all US federal spending. 'US citizens contribute about 17 cents per day to USAID, around US$64 per year,' said study co-author James Macinko of the University of California, Los Angeles. 'I think most people would support continued USAID funding if they knew just how effective such a small contribution can be to saving millions of lives.'

Struggling with math? Study shows a gentle electric jolt boosts arithmetic by up to 29pc — but only for some
WASHINGTON, July 2 — Struggle with math? A gentle jolt to the brain might help. A new study published yesterday in PLOS Biology suggests that mild electrical stimulation can boost arithmetic performance — and offers fresh insight into the brain mechanisms behind mathematical ability, along with a potential way to optimise learning. The findings could eventually help narrow cognitive gaps and help build a more intellectually equitable society, the authors argue. 'Different people have different brains, and their brains control a lot in their life,' said Roi Cohen Kadosh, a neuroscientist at the University of Surrey who led the research. 'We think about the environment — if you go to the right school, if you have the right teacher — but it's also our biology.' Cohen Kadosh and colleagues recruited 72 University of Oxford students, scanning their brains to measure connectivity between three key regions. Participants then tackled math problems that required either calculating answers or recalling memorised solutions. They found that stronger connections between the dorsolateral prefrontal cortex, which governs executive function, and the posterior parietal cortex, involved in memory, predicted better calculation performance. When the researchers applied a painless form of brain stimulation using electrode-fitted caps — a technique known as transcranial random noise stimulation — the low performers saw their scores jump by 25-29 per cent. The team believes the stimulation works by enhancing the excitability of neurons and interacting with GABA, a brain chemical that inhibits excessive activity — effectively compensating for weak neural connectivity in some participants. In fact, the stimulation helped underperformers reach or even surpass the scores of peers with naturally stronger brain wiring. But those who already performed well saw no benefit. 'Some people struggle with things, and if we can help their brain to fulfil their potential, we open them a lot of opportunities that otherwise would be closed,' said Cohen Kadosh, calling it an 'exciting time' for the field of brain stimulation research. Still, he flagged a key ethical concern: the risk that such technologies could become more available to those with financial means, widening — rather than closing — access gaps. He also urged the public not to try this at home. 'Some people struggle with learning, and if our research proves successful beyond the lab, we could help them fulfil their ambitions and unlock opportunities that might otherwise remain out of reach.' — AFP
