Ancient Egyptian man's genome reveals his society's cross-cultural ties

DNA obtained from the remains of a man who lived in ancient Egypt around the time the first pyramids were built is providing evidence of the ties between two great cultures of the period, with a fifth of his genetic ancestry traced to Mesopotamia.
Although based on a single genome, the findings offer unique insight into the genetic history of ancient Egyptians - a difficult task considering that Egypt's hot climate is not conducive to DNA preservation.
The researchers extracted DNA from the roots of two teeth, part of the man's skeletal remains that had been interred for millennia inside a large sealed ceramic vessel within a rock-cut tomb. They then managed to sequence his whole genome, a first for any person who lived in ancient Egypt.
The man lived roughly 4,500-4,800 years ago, the researchers said, around the beginning of a period of prosperity and stability called the Old Kingdom, known for the construction of immense pyramids as monumental pharaonic tombs.
The ceramic vessel was excavated in 1902 at a site called Nuwayrat near the village of Beni Hassan, approximately 170 miles (270 km) south of Cairo. The researchers said the man was about 60 years old when he died, and that aspects of his skeletal remains hinted at the possibility that he had worked as a potter.
The DNA showed that the man descended mostly from local populations, with about 80% of his ancestry traced to Egypt or adjacent parts of North Africa. But about 20% of his ancestry was traced to a region of the ancient Near East called the Fertile Crescent that included Mesopotamia.
"This suggests substantial genetic connections between ancient Egypt and the eastern Fertile Crescent," said population geneticist Adeline Morez Jacobs of Liverpool John Moores University in England and the Francis Crick Institute in London, lead author of the study published on Wednesday in the journal Nature.
The findings build on the archaeological evidence of trade and cultural exchanges between ancient Egypt and Mesopotamia, a region spanning modern-day Iraq and parts of Iran and Syria.
During the third millennium BC, Egypt and Mesopotamia were at the vanguard of human civilization, with achievements in writing, architecture, art, religion and technology.
Egypt showed cultural connections with Mesopotamia, based on some shared artistic motifs, architecture and imports like lapis lazuli, the blue semiprecious stone, the researchers said.
The pottery wheel from Mesopotamia first appeared in Egypt at about the time the man lived, a period when the earliest pyramids began to spring up near modern-day Cairo, starting with the Step Pyramid of the pharaoh Djoser at Saqqara and later the Great Pyramid of the pharaoh Khufu at Giza.
About 90% of the man's skeleton was preserved. He stood about 5-foot-3 (1.59 meters) tall, with a slender build. He also had conditions consistent with older age such as osteoporosis and osteoarthritis, as well as a large unhealed abscess from tooth infection.
"Ancient DNA recovery from Egyptian remains has been exceptionally challenging due to Egypt's hot climate that accelerates DNA degradation, with high temperatures breaking down genetic material over time compared to cooler, more stable environments," Francis Crick Institute population geneticist and study co-author Pontus Skoglund said.
"In this case, the individual's burial in a ceramic pot vessel within a rock-cut tomb likely contributed to the unusual DNA preservation for the region," Skoglund added.
The fact that his burial occurred before mummification became standard practice in Egypt may have helped avoid DNA degradation because his remains were spared elaborate preservation techniques.
Scientists have struggled to recover ancient Egyptian genomes, according to paleogeneticist and study co-author Linus Girdland Flink of the University of Aberdeen in Scotland. One previous effort yielded partial genome sequencing of three individuals who lived some 1,500 years after the Nuwayrat man.
Given the track record, the researchers were surprised with their success in sequencing the man's genome.
"Yeah, it was a long shot," Skoglund said.
The man may have worked as a potter or in a trade with similar movements because his bones had muscle markings from sitting for long periods with outstretched limbs.
"All indicators are consistent with movements and positions of a potter, as indicated in ancient Egyptian imagery," said bioarcheologist and study co-author Joel Irish. "He would have been of high status to have been buried in a rock-cut tomb. This conflicts with his hard physical life and conjecture that he was a potter, which would ordinarily have been working class. Perhaps he was an excellent potter." REUTERS

GEORGE TOWN - They can be found everywhere – airport lounge sofas, bus seats, and even hotel and hostel beds. But who would have known that bed bugs can play a role in forensic investigations? Scientists discovered that the blood they consume has the DNA of people whom they fed on. A Universiti Sains Malaysia (USM) team began studying the role of these blood suckers after collecting specimens at airport lounge sofas in 2014. Their study was published in Scientific Reports. USM researchers reported that Cimex hemipterus – a common tropical bed bug species – can retain human DNA in their guts for up to 45 days after feeding. In a subsequent research paper published in Forensic Science International in August 2024, the USM team outlined a practical field protocol for collecting and analysing tropical bed bugs as forensic evidence. Together, they extracted DNA from bed bugs, which had been fed human blood under controlled conditions. 'Right after feeding, we could recover full genetic profiles using STR (short tandem repeat) and SNP (single nucleotide polymorphisms) methods. 'And even 45 days later, we were still able to retrieve partial profiles – enough, in some cases, to point to hair, skin and eye colour,' Assoc Prof Hafiz said. The STR technique is the same one used in standard DNA profiling used in forensic labs. The research paper, Human profiling from STR and SNP analysis of tropical bed bug, Cimex hemipterus, for forensic science, marks the first documented forensic application involving this species. Previous research had only focused on Cimex lectularius, the common bed bug more prevalent in temperate climates. The discovery suggests that if bed bugs are found at a crime scene – a hotel room, a flat or a hideout – they may carry the genetic fingerprints of whoever was recently there. 'Bed bugs feed and then retreat into nearby crevices. 'Because they don't fly or travel far, there's a good chance that whoever they fed on had been at that exact location,' said Assoc Prof Hafiz. This makes them useful in crime scenes when conventional biological evidence – bloodstains or fingerprints – has been removed or cleaned up. He said even on day five, more than 70 per cent of the targeted STR readings were still intact, and 39 out of 41 SNP markers were readable, more than enough for partial profiling. He said though not yet admissible in court, this kind of DNA phenotyping could give investigators valuable leads when there are no other clues. The findings, while promising, still have limitations. The research used single-target polymerase chain reaction tests – a slower, one-at-a-time method – rather than the commercial multiplex kits used in accredited crime labs. Assoc Prof Hafiz said for bed bug DNA to be accepted in court, the protocols would need to be adapted and validated across different systems. There is also the issue of mixed DNA. 'Bugs can feed on more than one person, which can make it difficult to interpret the results,' he said. In field samples collected around Penang, the USM team often found more than two alleles – indicating the presence of more than one human profile. Assoc Prof Hafiz said following these discoveries, which had taken over 10 years, his team was ready to work with forensics experts on trial field applications to establish more detailed protocols. Meanwhile, Penang Crime Consciousness and Public Safety Society chairman Mohamad Anil Shah Abdullah said the findings could offer a new avenue in complex investigations. 'Sometimes, there's no blood or fluids at the scene. 'Insects like flies and mosquitoes have been used before but bed bugs don't fly. That makes them useful,' he said. He recalled a 2008 case in Finland where police retrieved a suspect's DNA from a mosquito in a stolen car. 'If a mosquito can do that, why not a bed bug?' he said. THE STAR/ ASIA NEWS NETWORK

The visitor from the stars, designated 3I/Atlas, is likely the largest yet detected, and has been classified as a comet, or cosmic snowball. 'It looks kind of fuzzy,' Dr Peter Veres, an astronomer with the International Astronomical Union's Minor Planet Centre, which was responsible for the official confirmation, said. 'It seems that there is some gas around it, and I think one or two telescopes reported a very short tail,' he added. Dr Richard Moissl, head of planetary defence at the European Space Agency, said the object, originally known as A11pl3Z before it was confirmed to be of interstellar origin, poses no threat to Earth. 'It will fly deep through the solar system, passing just inside the orbit of Mars', but will not hit our neighbouring planet, he said. This would mean it is not bound by the Sun's orbit, unlike objects that remain within the solar system. Its trajectory also 'means it's not orbiting our star, but coming from interstellar space and flying off to there again', Dr Moissl said. Dr Jonathan McDowell, an astronomer at the Harvard-Smithsonian Centre for Astrophysics, added: 'We think that probably these little ice balls get formed associated with star systems. 'And then as another star passes by, tugs on the ice ball, frees it out. It goes rogue, wanders through the galaxy, and now this one is just passing us.' A Chile based observatory that is part of the Nasa-funded Atlas survey first discovered the object on July 1. Professional and amateur astronomers across the world then searched through past telescope data, tracing its trajectory back to at least June 14. The object is currently estimated to be roughly 10km to 20km wide, Dr Moissl said, which would make it the largest interstellar interloper ever detected. But the object could be smaller if it is made out of ice, which reflects more light. Dr Veres said the object will continue to brighten as it nears the Sun, bending slightly under the pull of gravity, and is expected to reach its closest point – perihelion – on Oct 29. It will then recede and exit the solar system over the next few years. Our third visitor This marks only the third time humanity has detected an object entering the solar system from the stars. The first, Oumuamua, was discovered in 2017. It was so strange that at least one prominent scientist became convinced it was an alien vessel, though this has since been contradicted by further research. Our second interstellar visitor, 2I/Borisov, was spotted in 2019. There is no reason to suspect an artificial origin for 3I/Atlas, but teams around the world are now racing to answer key questions about things like its shape, composition and rotation. Dr Mark Norris, an astronomer at Britain's University of Central Lancashire, said the new object appears to be 'moving considerably faster than the other two extra-solar objects that we previously discovered'. The object is currently roughly around the distance from Jupiter away from Earth, Dr Norris said. He pointed to modelling estimating that there could be as many as 10,000 interstellar objects drifting through the solar system at any given time, though most would be smaller than the newly discovered object. If true, this suggests that the newly online Vera C. Rubin Observatory in Chile could soon be finding these dim interstellar visitors every month, Dr Norris said. Dr Moissl said it is not feasible to send a mission into space to intercept the new object. Still, these visitors offer scientists a rare chance to study something outside of our solar system.
