
EXCLUSIVE Experts finally find treatment for incurable viral infection suffered by 122million Americans
Half of Americans are suffering from an incurable disease that causes painful - and embarrassing - blisters and sores around the mouth.
About 122million people have herpes simplex virus 1 (HSV-1), the oral strain of herpes. It's spread though close skin-to-skin contact and differs from the sexually transmitted herpes simplex virus 2 (HSV-2), which causes genital blisters and sores.
There is no cure for HSV-1, but antiviral medications can manage outbreaks and reduce the risk of transmission.
Now, however, researchers from Spain, have discovered how the virus acts on a person's DNA, opening the door for a possible cure.
The team found that the virus 'hijacks' a certain enzyme in the body that allows herpes to replicate itself, but if that enzyme is blocked, it brings 'the hostile takeover to a halt.'
The researchers are hopeful that the study's findings, which provide the first proof herpes actually reshapes a person's DNA within only hours of infection, may help address the public health burden of HSV-1.
Globally, nearly 4billion people have this type of herpes and experts are becoming more concerned as drug-resistant strains are developing, which could lead to more transmission.
And unmanaged herpes can lead to devastating complications, traveling to the brain and triggering inflammation, which can increase the risk of dementia.
Researchers in Spain analyzed human cells and infected them with HSV-1, and found almost immediately that it reshapes cell DNA so it can access more genes and spread rapidly throughout the body.
However, blocking the enzyme topoisomerase I, which controls DNA replication, stopped HSV-1 from rearranging genes during infection, stopping it from spreading.
Blocking the enzyme can be done with drugs called topoisomerase inhibitors, which are usually used in chemotherapy regimens to stop DNA from replicating and forming more cancer cells.
This could be the first-ever method of slowing the spread of HSV-1.
The researchers said while more evidence supporting a possible treatment is needed, the study could be the first step in preventing worldwide herpes outbreaks.
Professor Pia Cosma, corresponding study author and researcher at the Centre for Genomic Regulation (CRG) in Barcelona, said: 'In cell culture, inhibiting this enzyme stopped the infection before the virus could make a single new particle.
'That gives us a potential new therapeutic target to stop infection.'
Herpes is most commonly transmitted from a carrier to a person without herpes by touching a cold sore, which actively produces or 'sheds' the virus.
However, it can cause genital herpes by spreading through oral sex.
HSV-1 leads to painful blisters around the lips and mouth, skin and genitals.
When the virus infects a person, it may travel up to a cluster of sensory nerves in the brain and remain dormant there for months or even years after the initial infection.
But in times of stress, severe fatigue, or changes to the immune system, the virus can reactivate, multiply, and travel back to the skin through nerve fibers. These stressful times can result in new blisters in the same area as the initial infection.
The new study, published Thursday in Nature Communications, looked at human A549 cells, which are caused by the cancer lung carcinoma. The cells were then infected with HSV-1 representing one, three and eight hours post infection.
Researchers found after eight hours, HSV-1 had occupied 70 percent of the cells, suggesting it would take less than a day for the virus to completely overtake cell DNA and spread.
Dr Esther Gonzalez Almela, first study author, said: 'HSV-1 is an opportunistic interior designer, reshaping the human genome with great precision and choosing which bits it comes into contact with.
'It's a novel mechanism of manipulation we didn't know the virus had to exploit host resources.'
The researchers then tried to suppress topoisomerase I, which relaxes DNA and makes it easier for it to replicate. They found this 'hindered viral replication.'
The team wrote that suppressing the enzyme stops HSV-1 from progressing, suggesting it could be most beneficial for those in later stages of infection.
Topoisomerase inhibitors are sold under names like etoposide, irinotecan and topotecan to slow the growth of lung, colorectal, ovarian and testicular cancers, among others.
Some are also used to treat multiple sclerosis, a progressive neurological disorder that attacks the spinal cord, by reducing central nervous system inflammation.
They can be given as either pills or intravenously for anywhere from $8 to $61 depending on the method.

Dentist cracks centuries-old maths puzzle hidden in famous Leonardo Da Vinci drawing
A London-based dentist has uncovered a hidden detail in Leonardo da Vinci 's famous Vitruvian Man drawing, revealing a special number connecting the iconic artwork to the human body and nature. The discovery, described in the Journal of Mathematics and the Arts, reveals how the legendary Italian polymath perfectly placed the Vitruvian Man human figure inside a circle and a square. An equilateral triangle 'hidden in plain sight' within Leonardo's notes could be a clue to his construction method, which laid the foundation for the drawing's proportional choice, says Rory Mac Sweeney, dentist and author of the study. The triangle can be found between the man's legs in the artwork, and 'isn't just a random shape', Dr Sweeney says. This shape matches one found in modern anatomy known as "Bonwill's triangle", which explains how human jaws work in the most efficient way, he says. The imaginary equilateral triangle is formed by connecting the centres of the points where the lower jaw connects to the skull, and the midpoint of the mandible's central incisors. The length of each side of Bonwill's triangle is generally equal in most individuals, at about 4 inches or 10 cm, and is used in dentistry to understand and analyse the anatomy and mechanics of the jaws. This measure helps design proportional dentures and position them for proper bite alignment. The use of such a triangle in Leonardo's art suggests the Italian polymath likely understood this ideal design of the human body centuries before modern science, Dr Sweeney suspects. Using such an equilateral triangle in the Vitruvian Man drawing helps produce a specific ratio – 1.64 – between the size of the square and the circle in the artwork. This ratio is almost identical to a "special blueprint number" 1.633, which appears frequently in nature for building the most efficient structures, such as the atomic structure of super-strong crystals, and the tightest way to pack spheres, like stacking oranges at the supermarket. 'Leonardo's systematic construction yields a ratio of 1.64 to 1.65 between the square's side and circle's radius, matching both published measurements of the original drawing, and the tetrahedral ratio of 1.633 found in optimal spatial arrangements,' the study noted. "We've all been looking for a complicated answer, but the key was in Leonardo's own words. He was pointing to this triangle all along," Dr Sweeney said. The discovery hints that the Vitruvian Man painting is not just a beautiful piece of art, but also a scientific work centuries ahead of its time. 'What's truly amazing is that this one drawing encapsulates a universal rule of design. It shows that the same 'blueprint' nature uses for efficient design is at work in the ideal human body,' Dr Sweeney said. 'Leonardo knew, or sensed, that our bodies are built with the same mathematical elegance as the universe around us,' he added.

Little-known signs of world's most infectious disease EVERYONE must know - as pandemic health chief issues chilling warning
The warning signs of measles have been highlighted as a top scientist has raised the alarm on declining vaccination rates. Science minister Lord Vallance of Balham warned that parents had forgotten just how deadly the world most contagious infection was before the invention of vaccines. 'People have forgotten that measles kills. They think, "Oh, measles—that's a pretty benign disease". It's benign because we've got vaccination', Lord Vallance told The Times. The 65-year-old added that to see people—most often children—die from entirely preventable disease was 'tragic'. 'People used to have 12 children because they knew five were going to die. It just doesn't happen like that anymore and vaccination has been a really major, major part of it,' he said. Measles usually starts with flu-like symptoms, including fever, cough and runny nose and is often followed by a tell-tale rash which starts on the face and behind the ears before spreading to the rest of the body. But it can trigger more serious and even fatal health complications if it spreads to the lungs or the brain. The incredibly contagious disease can infect 9 in 10 unvaccinated children in a classroom if just one classmate is infectious. One in five children who catch the disease will be hospitalised, according to estimates, with one in 15 developing life-threatening complications like meningitis or sepsis. Lord Vallance, who became a household name during the Covid pandemic, was speaking in the wake of a surge in measles in England. Measles cases soared in 2024 with 2,911 cases confirmed in England—the highest number on record since 2012—and tragically one death recorded. Despite surging rates of infections uptake of a vaccine designed to protect children against the disease has fallen. Less than 84 per cent of five-year-olds in the nation had both recommended doses of the measles, mumps and rubella (MMR) jab last year. Experts have labelled the figures concerning and urged parents to vaccinate their children to avoid the 'life-long consequences' of potential measles infection. But it's not just the MMR jab. Lord Vallance's warning comes as data shows none of the 14 childhood vaccines offered on the NHS met the 95 per cent uptake target needed for herd immunity. Herd immunity is a public health concept where enough people are vaccinated that it stops diseases spreading rampantly through the population as a whole. Experts have blamed post-pandemic scepticism and vaccine fatigue for the rapidly declining rates, with Vallance also pointing the finger at, now thoroughly debunked, rumours linking the MMR jab to autism for causing 'huge problems around the world'. Now, together with health chiefs, he is urging parents to get their children vaccinated—with two doses of the MMR jab offering up to 99 per cent protection. It comes as concerning figures released last week revealed nearly half of children in some parts of England haven't had both MMR jabs by the time they turn five. Data shows just 52.7 per cent of youngsters that age in Kensington and Chelsea in west London are fully vaccinated. London remains one of the least vaccinated parts of the country—taking up a staggering 19 of the top 20 spots for low MMR jab uptake. Outside of London, Nottingham was worst with just 71.4 per cent of children jabbed. Dr Doug Brown, Chief Executive of the British Society for Immunology, told MailOnline: 'Vaccination is the safest and most effective way to protect your child against measles. 'Measles is a serious disease that can make children very ill and cause life-long consequences. 'In the 2024 measles outbreak in England, 1 in 5 children who contracted the disease needed to be admitted to hospital for treatment. 'To be fully protected against contracting measles, it is essential that children receive two doses of the MMR vaccine at the correct timepoints. 'We need to redouble efforts to implement the NHS Vaccine Strategy and engage with communities to ensure that all children receive the MMR vaccine and are protected against these nasty diseases. 'It is never too late to get vaccinated—if you're concerned that your child has missed out on a vaccination, contact your GP surgery.' Lord Vallance became a household name during the pandemic, appearing next to Boris Johnson and his esteemed colleague Sir Chris Whitty during tense Downing Street briefings to talk the nation through the crisis. But he also earned the nicknamed 'Dr Doom' during the crisis for being the face of bleak projections from scientists.
