Orange Cats Have Long Been a Genetic Mystery. Scientists Have Finally Solved It

Cats with orange coats get them from a deletion (meaning a missing segment) mutation in the noncoding region of a gene associated with both neuroendocrine tissues and pigmentation.
There are more male orange cats than females because this gene is located on the X-chromosome, which males only have one copy of. Females are more likely to be tortoiseshell or calico because they have two X chromosomes, and therefore have more genetic coat color options to 'choose' from.
Garfield might be the most iconic orange tabby around, but Hollywood has seen quite a few leading cats of the same color—Heathcliff, Oliver, Hiyao Miyazaki's adorably terrifying Catbus from My Neighbor Totoro, and Morris from those retro cat food commercials all come to mind. Like most celebrities, they have so far refused to give up their most guarded beauty secret—how did they get those fabulous golden auburn coats?
Hiroyuki Sasaki—a cat enthusiast and geneticist at Kyushu University in Japan—was determined to identify the elusive gene that carries the orange mutation in Felis silvestris catus (the domesticated cat). He and his research team analyzed DNA in skin tissue from cats with and without orange fur, and found a mutation to the ARHGAP36 gene. This was a type of deletion mutation, meaning that a segment of a gene is missing. In orange cats, the missing segment is located on an intron, or noncoding region, of the ARHGAP36 gene, which is also in neuroendocrine tissues (especially the hypothalamus), adrenal glands, and pituitary glands.
The location of the mutation on this particular gene also explains why there are so many more male orange cats than female ones. ARHGAP36 is known as X-linked, meaning that it is located on the X chromosome. In female cats (and all female mammals), one of the two X chromosomes in each cell is randomly switched off in a process known as X chromosome inactivation, so even if the mutation is present, it is unlikely that it will be expressed by every cell and appear as an even (or even semi-even) orange coat. To be orange, a female cat must have the orange gene on both X chromosomes, so no matter which one is deleted, the orange gene still dominates.
Male cats, on the other hand, only have one X chromosome, and are therefore much more likely to evenly express that mutation.
Whether orange or not, all fur pigmentation genes are X-linked. Calico and tortoiseshell coats also come from different combinations of activated X chromosomes—both with and without the deletion that results in orange—which explains why most of them are female. For this to happen to a male cat, there would have to be two X chromosomes present next to the Y chromosome in order for random inactivation to result in mottled fur.
Sasaki and his team found that ARHGAP36 was most active in melanocytes (cells in the skin which produce pigment) found in the orange patches of calicos and tortoiseshells. Genes promoting melanogenesis, or the production of melanin in melanocytes, suppress ARHGAP36 and are upregulated in brown, black, and gray patches. These colors are associated with the black or brown pigment known as eumelanin, which is also the most common form of melanin.
Sasaki believes that when a mutated ARHGAP36 is expressed as orange fur in cats, the missing part of the gene would have suppressed orange coloration had it been present. In cats, mutated ARHGAP36 was shown to suppress other genes involved in the production of eumelanin so that it could instead produce a different type of melanin called pheomelanin, which is the reddish-yellow pigment in orange fur. Found only in mammals and birds, pheomelanin is also behind red hair in humans and flashy red feathers in some bird species.
It seems that high ARHGAP36 activity is, in general, associated with reduced activity in genes involved with the production of eumelanin. Sasaki is convinced that this gene's takeover may somehow shift pigment production to pheomelanin instead—though, how exactly it pulls this off is still unknown. And because ARHGAP36 also has significant importance in the brain, there is even speculation as to whether there are true associations between fur color and personality (the confirmation of which could either prove or silence all the orange cat memes out there).
'Although it is not fully understood how the identified deletion switches the pigment species, the variation likely dominates the cat population with orange coat color,' the researchers wrote.
Somewhere, Garfield is smugly beaming next to a tray of lasagna.

Here's what you'll learn when you read this story: Although CERN's Large Hadron Collider has made a lasting impact on particle physics, it hasn't yet open up a whole new frontier of particle physics like some scientists expected. One scientist champions a theory that new physics could be hiding in what he calls the 'zeptouniverse'—the realm of objects on the scale of the zeptometer (which is 1 quintillionth of a meter)—and that the best way to explore that universe is through observing kaon and B meson decays. Future colliders will likely be able to directly observe the zeptouniverse, but for now, studying these decays could help us find new physics within the decade. We humans have gotten pretty good at glimpsing the invisible. The Large Hadron Collider—our premier instrument for exploring the subatomic—can thoroughly explore the world of the attometer, which is (incredibly) just one-quintillionth of a meter. Famously, the LHC confirmed the existence of the Higgs boson in 2012, and physicists prepared for a rush of new particles to explain lingering mysteries of the universe like the existence of dark matter and matter-antimatter asymmetry. But that explosion of discovery didn't really materialize. That's certainly not to say nothing has happened since then, but no major revelations on par with the Higgs have been discovered since. And now, a new article in New Scientist, written by particle physicist Harry Cliff who works on the LHCb experiment, details one theory as to why we haven't found what we were expecting to find. At its most basic, many of these revelations could be hiding in what's sometimes referred to as the 'zeptouniverse,' which is a world that only exists at the 10-21-meter scale. The LHC can only analyze particles directly down to 50 zeptometers, but Cliff highlights a theory—largely championed by Technical University of Munich theoretical physicist Andrzej Buras—that these elusive particles could simply be beyond LHC's detection capabilities. Of course, a better detector could open up this frontier—CERN completed a feasibility study for the Future Circular Collider (FCC) just earlier this year. But Buras believes that we can explore this frontier of new physics indirectly without the need to wait the several decades required to finally probe this question (the FCC won't perform high-energy physics until 2070). In 2020, Buras explored this question in an article for Physik Journal, writing: Can we reach the Zeptouniverse, i.e., a resolution as high as 10–21m or energies as large as 200 TeV, by means of quark flavour physics and lepton flavour violating processes in this decade well before this will be possible by means of any collider built in this century? In a paper uploaded to the preprint server arXiv last year, Buras identified seven possible targets for this investigation, which he dubbed the 'magnificent seven,' according to New Scientist. All seven are extremely rare decays of particles containing strange and bottom quarks, which Cliff calls 'echoes from the zeptouniverse.' Luckily for Buras, some experiments are already searching for these ultra-rare decays. One example of such a decay starts with the B meson—a kind of composite particle made of different quarks, as Cliff explains. In 2023, the Belle II experiment in Japan captured this decay in action, producing another particle called a kaon (or K meson) and two neutrinos. However, because the experiment wasn't set-up to directly detect neutrinos, information about them is limited. This isn't the only ultra-rare decay that's been detected recently, either. In September of 2024, the NA62 experiment at CERN recorded the decay of a positively charged kaon into a pion and a matter-antimatter pair. It's thought that less than one in 10 billion kaons should decay in this way. Because this interaction is sensitive to Standard Model deviations, it's identified as one of the prime targets for finding new physics. Today, the KOTO experiment in Japan is searching for a second confirmation of this kaon decay. 'The search for new particles and forces beyond those of the Standard Model is strongly motivated by the need to explain dark matter, the huge range of particle masses from the tiny neutrino to the massive top quark, and the asymmetry between matter and antimatter that is responsible for our very existence,' Buras wrote last year in the trade magazine CERN Courier. 'As direct searches at the LHC have not yet provided any clue as to what these new particles and forces might be, indirect searches are growing in importance.' Scientists are only beginning to peer inside the unknown frontier of the zeptouniverse, and until next-generation colliders are up and running, these extremely rare decays are our only windows into that universe. It's not just an essential macronutrient, but also the building blocks for muscle strength, metabolism, and so much more. Talk to almost any expert, and they'll agree. 'Protein is an essential nutrient for so many bodily processes,' says Jessica Cording, RD, CDN, author of The Little Book of Game-Changers. 'It nurtures hair, skin, and nail health, builds and maintains tissues like muscles, supports connective tissue, and helps us feel full—and that's just scratching the surface.' There are, however, a *lot* of opinions out there on how much protein you should be getting each day. Your favorite fitness influencer may tout one number while your health-minded best friend tells you something else. The truth is, everyone's needs are slightly different. And if you're super active, that influences your number too. Want some protein inspiration in your life? Generally, most people require about 25 to 30 grams of protein per meal. It's a good idea to spread it out throughout the day, too, says Keri Gans, RDN, author of The Small Change Diet. Most people tend to pack in the protein at dinnertime, but in reality, you can (and should) eat it all day long because your body doesn't store protein. It breaks the macronutrient down into amino acids, which are used to build and repair tissues or to be used as fuel, so you need to keep the protein intake coming. Even knowing the general guideposts for protein intake, it can be hard to tell if it's the right number for you. You might find yourself wondering if you're hitting the gold standard on your protein intake or falling short. Ahead, experts break down the major signs that you might need more of this macronutrient in your life. Meet the experts: Jessica Cording, RD, CDN, is author of The Little Book of Game-Changers; Keri Gans, RDN, is author of The Small Change Diet; Gary Goldenberg, MD, is assistant clinical professor of dermatology at Icahn School of Medicine at Mount Sinai Hospital. 1) You're always hungry Say you just ate a meal, but you feel like you need a snack already. Cording says this is one of the most common signs that you're not having enough protein. Protein helps you to feel full, and if you're not getting enough, you may end up hungry again pretty soon after you eat. 'If someone is constantly hungry, I'm always going to check to make sure they have enough protein,' Cording says. 2) Your cuts and scrapes take a while to heal Protein is essential for wound-healing because it provides the basis for building new tissue and collagen, which are needed for your body's repair and growth, Cording explains. 'Sometimes when you don't eat enough protein, your body doesn't repair as well,' she says. 3) You're not building muscle Yep, even if you're going hard on the weights, a diet that lacks protein means you won't really see much improvement in muscle mass. Of course, you won't build muscle by eating protein alone, but if you're running into this issue, Cording says to reassess your protein. If you struggle to meet their daily protein needs within your regular meals, you can also opt for a protein powder to give yourself a little boost. But if you feel like there's no obvious reason for your hair loss, protein could be a culprit, according to Gary Goldenberg, MD, assistant clinical professor of dermatology at Icahn School of Medicine at Mount Sinai Hospital. 'Hair follicles are constantly cycling through growth (anagen), transition (catagen), and resting (telogen) phases,' he explains. 'Proteins are needed to support this constant turn over.' If you don't have enough protein, new healthy anagen follicles can't be produced—and you can end up losing hair, Dr. Goldenberg says. Of course, upping your protein intake isn't always a magic solution. 'While in most [people] this process can be repaired by an increase in protein intake, in some patients it can become chronic and long term,' he says. 5) Your nails are brittle Nails are mostly made of keratin, which is a form of protein. When there's not enough protein in your diet, you can't make and produce enough keratin, leading to brittle nails, Goldenberg says. Cue the dry and fragile nails. Of course, calcium, vitamin C, biotin (vitamin B7), and vitamin E are key players here, too. So, you should try to incorporate foods such as nuts, vegetables, and eggs into your diet, to reap all the benefits of these key vitamins. How To Get Enough Protein In Your Life Trying to eat 25 to 30 grams of protein at every meal can feel a little overwhelming. That's why Cording recommends starting small if you suspect you're not having enough of the macronutrient right now. 'Increase it in an approachable way,' she says. 'That might mean having a slightly larger portion of Greek yogurt, adding cottage cheese to your scrambled eggs, or adding a scoop of collagen powder to your coffee. That can add up.' Supplementing your diet with a protein powder shake can help. Gans recommends starting your day with protein to kick things off the right way. 'Don't skimp on protein at breakfast,' she says. Once you turn 40, you start to lose muscle mass and need more protein to stay strong and healthy, per the Mayo Clinic. Likewise, if you exercise regularly, you need more protein to meet your body's needs. Even if you don't fall into one of these categories, you should take steps to ensure you're packing enough protein. Protein deficiency can make you feel grumpy, sluggish, and hungry all the time—impacting everything from your weight to your mood. Plus, getting enough protein is easier said than done. Sarah Mirkin, R.D.N., author of Fill Your Plate, Lose The Weight, recommends getting at least 20 grams at each meal. Your body can only process 20 to 30 grams at once, she explains, so that's enough to keep you full and give you energy without going overboard. The thing is, one egg contains just 3.68 grams of protein and a slice of bacon offers just 2.58 grams, per the U.S. Department of Agriculture (USDA). So even if you think your meals are protein-packed, you may not be getting enough. Knowing the signs of protein deficiency can help you recognize when you're not meeting your body's needs. Here are the red flags that you need more protein on your fitness goals If you're working out but not seeing results, your post-workout meals may be to blame. That's because when you lift weights, your muscle fibers break down. And to help your body repair your muscles and make them stronger than before, you need a post-workout serving of protein. 'If you neglect to refuel with protein after your workouts, then you've only broken the muscle down, which results in muscle loss,' says sugar cravings Constantly craving sugar and carbs? When you eat a meal that lacks enough protein, you're more likely to feel unsatisfied and reach for sugary, fatty foods. 'Protein balances things out, stops cravings, and keeps us full longer,' says Natalie Jill, the lead trainer for Fit After 40. Be sure to consume enough protein at every meal to keep your sweet tooth in flaky skin If you used to have healthy, glowing skin that now looks dull and flaky, it might be a sign you're not getting enough protein. 'Protein makes up the building blocks of our hair, nails, and skin,' says Steven Bowers, D.O., author of Secrets of the World's Healthiest People. 'If you're skimping on protein, you may feel brain fog,' says Mirkin. 'Protein makes you feel more awake and alert, while carbohydrates can make you feel sleepy.' Iron deficiency, also known as anemia, can also cause extreme fatigue. Some foods high in iron also happen to be high in protein, so you can cover both bases by eating foods like beans, quinoa, ground beef, and chicken loss A thinner ponytail could be your body's way of crying out for help. 'Hair loss is very common from low-calorie and low-protein diets,' says Mirkin. 'Essential amino acids are necessary for healthy hair, skin, and nails.' So if your hair starts thinning or falling out, make sure you're getting enough protein. And if you are, talk to your doctor—hair loss is also associated with a range of other health that won't budge If you're losing weight, but your clothes are still tight, you may be dropping muscle instead of fat. This is especially true if you're not getting enough post-workout protein, or if you're over 40—the age when many people begin losing muscle mass. 'When our hormones start to change, eating protein becomes even more essential, because this is the time when our body starts losing lean mass,' says swings If you're feeling grumpier than usual, you may not be imagining it. Protein can boost your mood by providing the amino acids needed to produce feel-good neurotransmitters, like serotonin and dopamine. 'Without these essential amino acids, you are likely to experience depression, mood swings, and anxiety,' says or bloating If you constantly feel bloated—even while avoiding bloat-inducing beverages and foods—you may be low on protein. 'Protein is necessary to maintain an adequate balance of fluid in and outside of the cells of your body,' says Mirkin. And remember, bloating doesn't always happen in your belly. You can also experience swelling in your legs and ankles due to lack of immune system Taking more sick days than usual? A low-protein diet may be to blame, Mirkin says.
