‘Super humans' fears as scientists aim to create first human genome in a lab

Synthetic Human Genome (SynHG) project sparks fears that rogue scientists could create enhanced 'super humans' in decades to come
British scientists are launching a controversial project to create the first synthetic human DNA in a lab.
While they insist there is no intention to create a full synthetic human, researchers from universities of Oxford, Cambridge, Kent, Manchester and Imperial College London aim to write the first human genome from scratch. Some fear that opening this area of research could lead to rogue scientists seeking to create enhanced 'super humans' in decades to come. The Synthetic Human Genome (SynHG) project is being led by the Wellcome Trust, the world's largest medical research charity, and aims to boost health and treat incurable diseases.


Michael Dunn, research director at Wellcome said: 'Our DNA determines who we are and how our bodies work and with recent technological advances the SynHG project is at the forefront of one of the most exciting areas of scientific research. Through creating the necessary tools and methods to synthesise a human genome we will answer questions about our health and disease that we cannot even anticipate yet, in turn transforming our understanding of life and wellbeing.'
A leading genetic scientist has warned that creating human genetic material could lead to enhanced humans or even creatures that have human DNA.
Prof Bill Earnshaw, of Edinburgh University, told the BBC: "The genie is out of the bottle. We could have a set of restrictions now, but if an organisation who has access to appropriate machinery decided to start synthesising anything, I don't think we could stop them. If we manage to create synthetic body parts or even synthetic people, then who owns them. And who owns the data from these creations?"
How will it work?
The human genome is the genetic information that makes us physically who we are. It is made up of DNA which is contained in every cell in our body and carries the genetic information it needs to do its job.
Between 1990 and 2003 an international team of scientists took part in the Human Genome Project (HGP) to identify all human genes and sequence the three billion DNA base pairs. Now this information means scientists could take our understanding on another giant leap by not just reading DNA - but creating it.

The team behind it insist the work of the SynHG (Synthetic Human Genome) project will be confined to test tubes and petri dishes and there will be no attempt to create synthetic life. They will first build blocks of DNA to create a whole human chromosome which decide how our cells develop and repair themselves.
By experimenting on these chromosomes scientists could learn why some DNA reacts a certain way and how to modify it. This could help tackle inherited diseases which occur when genes go wrong.
Prof Matthew Hurles, director of the Wellcome Sanger Institute, said: "Building DNA from scratch allows us to test out how DNA really works and test out new theories, because currently we can only really do that by tweaking DNA in DNA that already exists in living systems".

The team hope to first create a fully synthetic human chromosome, which makes up approximately 2% of our total DNA.
Professor Jason Chin, of Oxford University, will lead the SynHG project. He said: 'The ability to synthesize large genomes, including genomes for human cells, may transform our understanding of genome biology and profoundly alter the horizons of biotechnology and medicine. With SynHG we are building the tools to make large genome synthesis a reality, and at the same time we are pro-actively engaging in the social, ethical, economic and policy questions that may arise as the tools and technologies advance.'

The Wellcome Trust cautions that building a complete synthetic human genome to help change and treat human health could take decades.
Prof Robin Lovell-Badge, of the Francis Crick Institute, said the project could create 'synthetic cells that can be grown in a lab'. He added: 'If these were to ever be used in humans, it would be important to design them carefully so that they can't lead to tumours or produce novel infectious particles. I would urge incorporating a genetic 'kill switch' to eliminate them from any location in the body or at least to make them easy for the immune system to find and destroy. There is no suggestion to make synthetic humans. We have no idea how to do this and it is likely to be very unsafe.'
Such research has been taboo until now because of concerns it could lead to designer babies or unforeseen changes for future generations.
Sarah Norcross, Director of the Progress Educational Trust (PET), said: 'We must recognise that this sort of work is not without controversy, and that is vital for researchers and the public to be in communication with one another. The public must have a clear understanding of what this research entails, while researchers and funders must have a thorough understanding of where the public wants to go with this science.'
Dr Tom Collins, research lead at Wellcome, told the BBC: "We asked ourselves what was the cost of inaction. This technology is going to be developed one day, so by doing it now we are at least trying to do it in as responsible a way as possible and to confront the ethical and moral questions in an upfront way as possible".

The Cave of the Crystals is one of the most stunning geological wonders at the centre of the Earth – but it's not a place you would want to stay for long because death would find you within minutes A remarkable geological marvel located at the heart of our planet has been dubbed the "best kept and most breathtaking secret on the planet" - but visiting it would be a deadly risk. The hostile underground cavern, known as the Cave of the Crystals, is situated in Chihuahua, Mexico. Broadcaster Michael Usher, who negotiated for two years to gain access, described it as "mother nature's Sistine Chapel" that is "almost something from out of space". The cave, filled with spectacular crystals weighing up to 55 tonnes and reaching 11 metres in height, lies 300 metres beneath a lead mine in the town of Naica. ‌ The crystal-filled chamber was only discovered in 2000 when miners stumbled upon it during drilling operations. The cave, shaped like a horseshoe, extends for 110 metres. Despite its mesmerising interior, lingering there too long could prove fatal. ‌ Recently, 60 Minutes Australia reissued a video showing their reporter being granted rare access, where he stated: "The furnace-like heat and soaring humidity can kill you in minutes. But when you see what I saw - you will see why it was worth every suffocating gasp for breath." Humans are not designed to survive in 58Cheat coupled with stifling humidity. Before Michael descended into the cave, an accompanying doctor warned: "This is going to be the harshest environment that you and I are ever going to come across in our life. So I am excited but a little anxious." The crystals were described as "the most spectacular anywhere in the word" with Michael saying he was "literally blown away" by the beauty of it. Also present was British geologist Dougal Jerram who marvelled at what he was seeing, saying it was the stuff of make believe while comparing it to something you would see in a Walt Disney production. However, despite the scenery, the trio were soon sweating profusely, comparing the feeling to like running a marathon. The humidity is at a perilous 90% and people aren't allowed to stay more than 10 minutes, even with specially-designed cooling suits. ‌ The doctor said they were essentially being cooked from the inside and the geologist added: "It is a remarkable place but it is so overpowering in terms of its attack on you in the environment. You are in this kind of constant battle of being overpowered physically, mentally and psychologically in every way." Struggling for breath, they soon left the cave, and were left to reflect on what they witnessed. The crystal cave they explored was first formed some 26 million years ago when magma rose from the reservoir, which, according to Live Science, pushed mineral-rich waters up through cracks in the rock. This opened up caverns in the limestone before the deposits were crystalised over thousands of years. Following his visit to the "enormous" natural wonder, roughly the size of a football pitch, geologist Dougal was left in awe, stating: "Oh, this is truly one of the most... if we were to list seven geological wonders of the world, this is definitely one of them." Due to its perilous nature, scientists are required to obtain a special permit to explore the Cave of the Crystals, and it remains strictly off-limits to tourists.

EXCLUSIVE Breakthrough British prostate cancer test could avoid need for biopsies in 80pc of men

A British-made prostate cancer test could avoid the need for biopsies in four out of five cases, according to a study. The EpiSwitch PSE next generation blood test uses advances in technology to identify prostate cancer, the most common form of the disease among men in the UK. The test, made by Oxford BioDynamics, has an accuracy of 94 per cent in identifying prostate cancer, a dramatic leap compared with the standard test along, which is reckoned to be correct in only around half of cases. Crucially, a peer-reviewed study due to be published in the Cancers medical journal, accepts the findings that up to 80 per cent of patients could potentially avoid unnecessary prostate biopsies - an often invasive procedue considered one of the biggest stumbling blocks in men going to get tested in the first place. The study concludes: 'Across models, up to 79.1 per cent of patients could safely defer biopsy based on PSE results, highlighting the test's potential to reduce invasive procedures without compromising diagnostic safety.' Dr Alexandre Akoulitchev, chief scientific officer at Oxford BioDynamics, said: 'This study builds on our earlier robust clinical validation work which demonstrated the application of EpiSwitch PSE as a precise, minimally invasive test that empowers clinicians and patients with clarity, reduced patient risk, and improved outcomes, while easing the pressure on the diagnostic pathway.' The paper said the innovation could effectively save the NHS significant amounts of money by reducing the amount of men sent for biopsies and MRI scans, reducing pressure on the service. The study reckoned this translated to a saving of almost £1,000 per patient. Oliver Kemp, chief executive of UK charity Prostate Cancer Research (PCR), said: 'This study shows how smarter testing can save millions while improving care for patients. 'Reducing unnecessary biopsies and scans means less stress for men and more capacity in the system.' Oxford BioDynamics worked with leading researchers and clinicians at the Imperial NHS Trust, Imperial College London, and the University of East Anglia to produce EpiSwitch. It is used following a PSA test - meaning prostate-specific antigen, identifying whether the protein levels produced by the gland are raised - and measures five markers to identify the likelihood of cancer being present. The firm's headquarters were visited by former Prime Minister Rishi Sunak in February this year, as he backed calls for a targeted national screening programme for men most at risk of prostate cancer. In government, Mr Sunak announced with charity PCR a £42 million screening trial to find ways of detecting prostate cancer earlier. But it is hoped a national screening programme would be the most effective way to prevent unnecessary deaths. The Mail has long campaigned for a national roll-out, a cause backed by the likes of Sir Stephen Fry, a former prostate cancer patient. The UK national screening committee has come under increasing pressure to reconsider testing policy following the news last year of Sir Chris Hoy's diagnosis with the terminal illness. Prostate cancer most frequently affects black men and those with a family history. It is often symptomless, meaning early detection is crucial in stopping the disease's spread. Earlier this week, newscaster Dermot Murnaghan revealed he is battling stage-four prostate cancer.
